长链非编码RNA BC032469通过调控hTERT影响肺癌细胞的增殖、迁移及侵袭

Downregulation of long non-coding RNA BC032469 suppresses lung cancer cell proliferation, migration and invasion by regulating hTERT

目的:探讨长链非编码RNA(longnon-codingRNA,lnc RNA)BC032469在肺癌细胞系中的表达及对细胞增殖、迁移和侵袭的影响。方法:采用实时荧光定量PCR(quantitative real-time PCR,qRT-PCR)测定肺癌细胞系(H1437,A549,H1299,H1975及H460)和人支气管上皮细胞(human bronchialepithelialcells,HBE)中lncRNABC032469的表达;设计沉默lncRNABC032469的siRNA,利用qRT-PCR检测lncRNA BC032469 siRNA在肺癌细胞中的沉默效率;CCK-8法检测细胞增殖能力的变化;划痕和Transwell试验评估细胞的迁移和侵袭能力的变化;qRT-PCR及Western印迹法分别测定h TERT mRNA和蛋白水平表达的变化。结果:LncRNA BC032469在肺癌细胞系(H1437,A549,H1299,H1975及H460)中相对表达量高于HBE(P<0.05)。沉默lncRNABC032469后,可以抑制H460细胞的增殖、迁移和侵袭,差异有统计学意义(P<0.05)。结论:LncRNA BC032469在肺癌细胞系中高表达,沉默lncRNA BC032469抑制肺癌细胞的增殖、迁移和侵袭能力,其机制可能与下调h TERT表达有关,提示lncRNA BC032469可以作为临床肺癌治疗的潜在靶标。

Objective： To explore the expression of long non-coding RNA（lncRNA BC032469） in lung cancer cell lines and its effect on proliferation, migration and invasion. Methods： We compared the expression of lncRNA BC032469 in lung cancer cell lines（H1437, A549, H1299, H1975 and H460） and human bronchial epithelial cells（HBE） by quantitative real-time PCR（qRT-PCR）. qRT-PCR was used to detect the silence efficiency of lncRNA BC032469 silence sequence. The proliferation, migration and invasion ability of cells were assessed by CCK-8 assay, Scratch assay and Transwell assay after the lncRNA BC032469 siRNA transfection. qRT-PCR and Western blot were used to detect the expression of hTERT. Results： qRT-PCR results showed that lncRNA BC032469 expression was higher in lung cancer cells（P〈0.05）. Knockdown of lncRNA BC032469 inhibited proliferation, migration and invasion of H460 cells（P〈0.05）. Conclusion： LncRNA BC032469 is highly expressed in lung cancer cells. Knockdown of lncRNA BC032469 inhibits lung cancer cells proliferation, migration and invasion and the mechanism may be related to the down-regulated of hTERT, which indicating that lncRNA BC032469 may be a candidate target of lung cancer gene therapy.
