降香提取物B3对斑马鱼模型促血管新生作用的研究 被引量:5

摘要 目的:评价降香提取物B3的促新血管生成作用及其机理。方法:采用转基因斑马鱼模型,通过观察转基因斑马鱼肠下血管(SIVs)新生模型及节间血管(ISVs)损伤模型来评价降香提取物B3的促新血管生成作用及其机理。结果:降香提取物B3能剂量依赖性地促进转基因斑马鱼SIVs区域的毛细血管出芽新生,30μg/ml效果最佳;能修复VEGF受体激酶抑制剂Ⅱ(VRI)诱导的ISVs区域血管损伤,且能上调VEGF受体激酶抑制剂Ⅱ诱导后的kdr、kdrl及flt-1等VEGF受体mRNA水平的下降。结论:降香提取物B3能通过调控VEGF受体mRNA水平以发挥其在转基因斑马鱼上的促血管新生效果,具有被开发为治疗性血管新生药物的潜力。 Objective: To evaluate the Dalbergia extract B3 promoting angiogenesis and its mechanism. Methods: In the transgenic zebrafish,neonatal sub intestinal veins(SIVs) model and intersegmental vessels(ISVs) injury model were used to evaluate Dalbergia extract B3 promoting angiogenesis and its mechanism. Results: Extract B3 of Dalbergia dose-dependently promoted capillaries to sprout in the transgenic zebrafish SIVs region and 30 g/ml was the optimized dose. Extract B3 of Dalbergia repaired VEGF receptor kinase inhibitor(VRI) induced vascular damage in the ISVs region,and upregulated decreased levels of kdr,kdrl and Flt-1 mRNA. Conclusion: Extract B3 of Dalbergia could regulate VEGF receptor mRNA level in transgenic zebrafish to exert its angiogenic effect.
