大黄素和芹菜素对高血压大鼠的肾脏保护及抗氧化作用 被引量:20

Renal protection and antioxidant effect of emodin and apigenin on spontaneously hypertensive rats

摘要 目的:探讨大黄素、芹菜素及大黄素和芹菜素联合应用对自发性高血压大鼠(SHR)的降压和肾脏保护及抗氧化作用。方法:13周龄SPF级雄性SHR大鼠随机分成5组:模型组、大黄素16mg/kg组、芹菜素110mg/kg组、大黄素16mg/kg和芹菜素110mg/kg联合应用组及卡托普利30mg/kg组。以同周龄SPF级SD雄性正常大鼠(n=8)为正常对照组。各组大鼠每日灌胃相应药物和溶媒。给药前及给药6周后分别测定大鼠血压; 6周后,测定右肾质量指数;检测肾功能血浆肌酐(SCr)、尿素氮(BUN);测定肾脏超氧化物歧化酶(SOD)、丙二醛(MDA)及血管紧张素Ⅱ(AngⅡ)含量; HE、Masson染色观察肾组织病理变化。结果:各给药组均明显降低SHR大鼠血压,升高右肾质量指数,大黄素和芹菜素联合应用较单用药更明显;显著降低血浆SCr、BUN及肾脏MDA、AngⅡ含量,升高肾脏SOD水平,且联合应用更显著;大黄素和芹菜素及联合用药均能减轻肾间质纤维化,减少肾小球系膜区及肾小管胶原沉积。结论:大黄素和芹菜素对自发性高血压大鼠能降低收缩压,并具有部分肾脏保护作用,且联合用药更有优势,可能与其抗氧化应激反应有关。 Aim: To research the antihypertensive,protective and antioxidant effect of emodin and apigenin on kidney in spontaneously hypertensive rats(SHR). Methods: 13-week-old male SHR(n = 40) were randomly divided into five groups: model group(SHR),emodin group(16 mg/kg),apigenin group(110 mg/kg),emodin(16 mg/kg) and apigenin(110 mg/kg) group,captopril group(30 mg/kg). Another same week-old male sprague dawley(SD) rats(n = 8) were given 0. 4% CMC as controls. All animals were administrated intragastrically once a day,10 ml/Kg. The drugs were dissolved in 0. 4% CMC. The systolic blood pressure(SHR) was measured before and after the administration. After six weeks,the ratio of right kidney weight and body weight,blood plasma creatinine(SCr) and urea nitrogen(BUN),SOD,MDA and angiotensinⅡ(AngⅡ) in kidney were measured. The kidney was excised for hematoxylin and eosin(HE) and Masson stain to observe the changes of pathology. Results: The results showed that emodin and apigenin significantly decreased the systolic blood pressure(SBP) of SHR,attenuated the development of renal interstitial fibrosis with the collagen deposition of renal mesangial region and kidney tubules lessened as compared with model group. Furthermore,the contents of SCr,BUN,MDA and AngⅡ in drug groups were obviously decreased with SOD level increased(P 〈 0. 01). The ratio of right kidney weight and body weight in drug groups were increased(P 〈 0. 01). The combination of emodin and apigenin was more significant than that of alone(P 〈 0. 01). Conclusion: These findings indicate that emodin and apigenin can decrease SBP in SHR. The protective effect of emodin and apigenin on kidney in SHR may be related to increasing anti-oxidative stress. The combination of drugs is more advantageous.
