摘要
目的:观察双藤汤(昆明山海棠、黑骨藤)醇提取物对寒痹型胶原诱导关节炎大鼠抗炎作用的影响。方法:取雄性SD大鼠,除空白组外,其余动物采用胶原诱导关节炎模型。成模动物随机分为模型组、双藤汤60、40、20g生药/kg组、青藤碱0. 028g/kg组,每组10只。连续给药4周后测量动物足跖厚度; HE染色观察踝关节和脾脏病理变化; Western-blotting检测脾脏和关节滑膜IKB-α、TAB-2、IKK-α蛋白表达; ELISA检测细胞因子TNF-a、IL-6、IL-1β。结果:与模型组比较,空白组、双藤汤60g、40g生药/kg组和青藤碱0. 028g/kg组能够显著抑制足跖肿胀程度;脾脏和踝关节病理显示双藤汤60g、40g生药/kg组和青藤碱0. 028g/kg组抑制脾小体萎缩减少和保持踝关节面光滑完整,软骨细胞整齐排列; Western-blotting显示双藤汤60、40、20g生药/kg组、青藤碱0. 028g/kg组IKB-α、TAB-2、IKK-α蛋白表达量受到不同程度抑制; ELISA显示双藤汤60g、40g生药/kg组、青藤碱0. 028g/kg组TNF-α、IL-6、IL-1β表达量下降。结论:双藤汤可能通过抑制TNF-α、IL-6、IL-1β、IKB-α、TAB-2、IKK-α蛋白的过表达起到对寒痹型胶原诱导关节炎大鼠炎症的抗炎作用。
Objective: To observe anti-inflammatory effect of Shuangteng decoction on Collagen-Induced Arthritis rats aggravated by cold. Method:Male SD rats were randomly selected and divided into sham group,model group,60 g/kg,40 g/kg and 20 g/kg group of Shuangteng decoction and 0. 028 g/kg group of sinomenine,10 in each group. Collagen-Induced Arthritis was constructed in the other groups except for the sham group. After 4 weeks of continuous administration,the thickness of vola pedis were measured and the pathological changes were observed by Ankle joint and spleen HE staining. Western-blotting was used to detect the expression of IKB-α,TAB-2,IKK-αprotein in spleen and synovial membrane. ELISA was used to detect TNF-a,IL-6,IL-1β. Result: Compared with the model group,the swelling degree of vola pedis were inhibited in sham group,Shuangteng decoction 60 g/kg group and 40 g/kg group and sinomenine 0. 028 g/kg group. The spleen and ankle joint pathology showed decrease of spleen corpuscle atrophy were inhibited and the ankle joint surface were kept smooth and Chondrocytes were orderly arranged in sham group,Shuangteng decoction 60 g/kg and 40 g/kg group and sinomenine 0. 028 g/kg. Western-blotting showed that the expression of IKB-a,TAB-2 and IKK-a protein was inhibited in 60 g/kg,40 g/kg and 20 g/kg group of Shuangteng decoction and 0. 028 g/kg group of sinomenine respectively. ELISA showed that the expression including TNF-α,IL-6,IL-1βwas decreased in 60 g/kg and 40 g/kg group of Shuangteng decoction and 0. 028 g/kg group of sinomenine. Conclusion: Shuangteng decoction could inhibit the overexpression of TNF-a,IL-6,IL-1β,IKB-a,TAB-2,IKK-a protein and inhibit the inflammation of Collagen-Induced Arthritis rats aggravated by cold.
