补肾润肺口服液对小鼠精子质量的影响研究

Study on the effect of Bushen Runfei oral liquid on sperm quality in mice

目的:研究补肾润肺口服液对小鼠精子活力的影响,为探索新适应症提供实验依据。方法:参照《WHO人类精液及精子-宫颈黏液相互作用实验室检验手册》,采用小鼠体内实验法,观察不同剂量的补肾润肺口服液组对精子活力的影响,对比了阳性对照肾宝合剂组以及阴性对照组小鼠睾丸重量、附睾尾精子的平均路径速度、直线运动速度、曲线运动速度、精子头侧摆幅度、前向性、直线性、精子活率和精子活力、异种穿卵的受精率等。结果:补肾润肺口服液10、20、40ml/kg组的睾丸重量较阴性对照组和肾宝合剂组有显著升高。补肾润肺口服液10、20、40ml/kg组小鼠附睾尾精子的精子活率、精子活力(a+b)较阴性对照组显著升高。补肾润肺口服液10、20、40ml/kg组小鼠附睾尾精子的平均路径速度(VAP,um/s)、直线运动速度(VSL,um/s),曲线运动速度(VCL,um/s)、精子头侧摆幅度(ALH,um/s),鞭打频率(BCF,Hz),前向性(STR,%)和直线性(LIN,%)高于阴性对照组;其中,补肾润肺口服液低、中和高剂量组的VSL、VCL、VAP较阴性对照组显著升高。补肾润肺口服液低、中、高剂量组以及肾宝合剂阳性对照组动物异种穿卵的受精率与阴性对照组相比有显著提高。结论:补肾润肺口服液对小鼠附睾尾精子质量具有一定促进作用。

Objective： study the effect of Bushen Runfei oral liquid on sperm motility in mice to explore the new indications with experimental evidences. Methods： conduct the in vivo mice experiments according to the WHO laboratory manual for the examination and processing of human semen,Shenbao Heji group as positive control and water as negative control in the experiments,the mice testicle weight,sperm viability and motility,fertilization rate were compared together with epididymal sperm exercise index such as average path velocity（VAP,um/s）,linear velocity（VSL,um/s）,curvilinear velocity（VCL,um/s）,sperm head side swing scope（ALH,um/s）,forward（STR,%）,linear（LIN,%）. Results： The research showed the Bushen Runfei oral liquid had significantly higher testicle weight than negative control and positive control groups（P 〈 0. 01）. The sperm viability and motility（a ＋ b） were better than negative control（P 〈 0. 05） for Bushen Runfei oral liquid with low,medium and high dose groups,also with higher epididymal sperm exercise index than negative control,especially,the VCL of low and medium Bushen Runfei oral liquid dose groups were significantly increased（P 〈 0. 05）. The fertilization rate for three Bushen Runfei oral liquid dose groups and positive control were higher than negative control（P 〈 0. 05）. Conclusion： These results indicated the Bushen Runfei Oral Liquid had a certain role of promoting on the sperm quality in mice.
