摘要
目的探讨腺苷A_(2A)受体激动剂CGS21680对激活的原代小胶质细胞肿瘤坏死因子-α(TNF-α)表达的影响。方法采用新生小鼠大脑皮质小胶质细胞原代培养技术,将培养的原代小胶质细胞随机分为3组。(1)对照组:空白对照;(2)脂多糖(LPS)组:诱导小胶质细胞激活,制作小胶质细胞炎症细胞模型;(3) CGS21680处理组:小胶质细胞炎症细胞模型基础上叠加CGS21680处理,CGS21680处理组按CGS21680不同浓度分为0.01、0.1、1和10μmol·L^(-1)4个亚组。采用ELISA法检测各组TNF-α含量。结果 LPS孵育24 h后,与LPS组比较,CGS21680 0.1μmol·L^(-1)处理组、CGS21680 1μmol·L^(-1)处理组和CGS21680 10μmol·L^(-1)处理组的TNF-α表达水平显著降低(均P<0.001)。与LPS组比较,CGS21680 10μmol·L^(-1)处理组的12、24、48和72 h时间点TNF-α表达水平均显著下降(抑制作用),呈显著的干预作用和时间-效应关系(均P<0.05)。结论腺苷A_(2A)受体激动剂CGS21680可明显抑制激活的小胶质细胞的炎症反应。
Aim To investigate the effect of CGS21680, an adenosine A2A receptor agonist, on the expression of TNF-α in activated microglia cells. Methods Primary murine microglia cells were cultured and randomly divided into three groups: a control group, a LPS group(established inflammation model in vitro), a CGS21680 treated group(inflammation model in combination with CGS21680 treatment). The CGS21680 treated group was further divided into 0.01, 0.1, 1 and 10 μmol·L^(-1) CGS21680 subgroups according to different concentrations of CGS21680. The expression of TNF-α by microglia was detected by ELISA. Results The expression of TNF-α was reduced in CGS21680 treated group compared with LPS group. Conclusion Adenosine A2A receptor agonist CGS21680 inhibits the expression of TNF-α in activated microglia cells.
