摘要
目的:(1)研究Livin基因在喉癌组织中的表达;(2)设计siRNA沉默喉鳞癌细胞Livin基因,检测其细胞中Livin蛋白表达。方法:(1)采用免疫组织化学MaxVision法染色检测87例喉癌组织,79例癌旁组织及19例转移淋巴结中Livin蛋白的表达情况;(2)用Western blot对SH2-R、SH2-F、NC组的喉鳞癌细胞(hep-2)的Livin蛋白进行分析。结果:(1)喉鳞癌组织中Livin表达阳性率为71. 26%,显著高于癌旁喉组织。TNM分期中Ⅲ+Ⅳ期喉癌阳性表达率为89. 28%,明显高于TNM分期中Ⅰ+Ⅱ期喉癌;在喉鳞癌颈淋巴结转移阳性者Livin阳性表达率高达73. 68%,高于颈淋巴结转移阴性者。(2) SH2-R、SH2-F、NC组的Livin蛋白相对表达量以SH2-R组最低(12. 31±4. 43)%,SH2-R组与SH2-F、NC组比较有统计学意义。结论:Livin基因在喉癌组织中高表达;在体外,siRNA降低了喉鳞状细胞癌中Livin的表达。
Objective: To study the expression of Livin gene in laryngeal carcinoma.Design the Livin gene of siRNA in laryngeal squamous cell carcinoma and detect the expression of Livin protein in the cells. Methods: Immunohistochemical MaxVision staining was used to detect the expression of Livin protein in 87 cases of laryngeal carcinoma,79 cases of paracancer tissue and 19 cases of metastatic lymph nodes.Western blot were used to analyze the Livin protein of SH2-R,SH2-F and NC in laryngeal squamous cell carcinoma. Results: The positive rate of Livin expression in the tissues of laryngeal squamous cell carcinoma was 71.26%,which was significantly higher than that of the paracancerous larynx.The positive expression rate of clinical stage Ⅲ+Ⅳ was 89.28%,which was significantly higher than that of stage Ⅰ+Ⅱ.In the metastasis of cervical lymph nodes,the positive expression rate of Livin was 73.68%,which was higher than those with negative cervical lymph node metastasis.The lowest relative expression of Livin protein in group SH2-R,SH2-F and NC was the SH2-R group,and the result was (12.31±4.43)%.There was a significant statistical difference between group SH2-R and group SH2-F and NC. Conclusion: Livin gene is highly expressed in larynx tissue,and in vitro,siRNA reduces the expression of Livin in laryngeal squamous cell carcinoma.
