银屑病易感基因的生物信息分析

摘要 目的分析银屑病易感基因的功能,筛选关键基因。方法结合OMIM数据库以及近年来发表的文章收集银屑病易感基因,利用Gather在线服务对易感基因进行富集分析,蛋白质相互作用网络由String数据库构建,计算节点的度数,据此筛选关键易感基因。结果银屑病的易感基因功能主要富集在免疫应答,防御反应,生物应激反应,有机生理过程,应激反应,对害虫寄生虫病原体的反应,对外部生物刺激的反应,对外伤的反应,抗原表达,抗原呈递及抗原加工等类别。我们通过计算易感基因所编码的蛋白质相互作用网络中节点的度数,按度数的高低,选出排在前十的10个基因,作为关键易感基因,功能分析和通路分析提示关键基因和很多功能明确与银屑病相关的基因相互作用。结论易感基因分析提示,银屑病是与环境因素密切相关,具有遗传性的免疫性皮肤病。蛋白质相互作用网络分析提示,关键易感基因可以没有明确的功能,但是会和很多与银屑病相关功能明确的基因相互作用。 Objective To analyze the functional enrichment of psoriasis susceptible genes and screen the key susceptible genes. Methods According to the OMIM database and the published articles in recent years,the susceptibility genes of psoriasis were collected,and the susceptibility genes were enriched and analyzed by Gather online service. The protein interaction network was constructed by using the String database, and the key susceptible genes were screened. Results The function of susceptibility gene of psoriasis is mainly enriched in immune response, defense response, biological stress response, organic physiological process, stress response, response to insect parasite pathogens, response to external biological stimuli, response to trauma, antigen expression, antigen presentation and antigen processing. By calculating the degree of the nodes in the protein interaction network encoded by the susceptibility genes, we selected the first 10 genes, as the key susceptible genes. Both the functional analysis and pathway analysis suggested that the key genes interact with the many genes related to psoriasis. Conclusion Susceptibility gene analysis indicates that psoriasis is a hereditary immune skin disease, closely related to environmental factors. Protein interaction network analysis suggests that though some key susceptible genes do not have definite functions, but they interact with a lot of genes with specific functions associated with psoriasis.
