摘要
【目的】为了建立番茄植株中茉莉酸的毛细管区带电泳分析新方法。【方法】详细研究了影响茉莉酸分离的因素如分离缓冲体系、pH、样品中有机溶剂含量等。番茄植株在液氮中研碎,用40%甲醇提取,离心除杂后,以50μm i.d.%40.2cm (有效长度30cm)未涂层熔融石英毛细管为分离柱,以20mmol/L硼砂(pH9.20)为分离缓冲液。检测波长为200nm、分离电压为12kV、进样时间为5s。【结果】在20.0、40.0、80mg/L三个添加水平下,方法的回收率为93.2%~102.3%之间,相对标准偏差(n=5)小于2%。【结论】本方法为毛细管检测痕量茉莉酸提供了分离依据。
【Objective】A new analyzing method of jasmonic acid in tomato plants was investigated by capillary zone electrophoresis.【Methods】The extraction conditions of jasmonic acid such as extraction buffer system, pH, organic solvent content in sample were studied. Tomato plants were grinded in liquid nitrogen, and extracted by 40% methanol, then centrifugated. The analysis was carried out using an uncoated capillary with 50 mm i.d and 40.2 cm (effective length: 30 cm) total length. The running buffer was 20 mmol/L Na 2 B 4 O 7 (pH 9.20) solution. The operation voltage was 12 kV. The detection wavelength was set at 200 nm. The injection time was 5 s at 0.5 psi. 【Results】The average recoveries at the three spiked levels (20, 40 and 80 mg/L) were from 93.2% to 102.3% with relative standard deviations all lower than 2%.【conclusion】The present method provided a valuable asset for investigation of trace jasmonic acid by capillary electrophoresis.
