摘要
目的建立桂附地黄丸(浓缩丸)中莫诺苷和马钱苷的总量与丹皮酚含量的测定方法。方法采用高效液相色谱法测定方中酒萸肉中莫诺苷和马钱苷的总量与牡丹皮中丹皮酚的含量;色谱柱为phenomsil C_(18)(250mm×4. 6mm,5μm),流动相:以乙腈为流动相A,以0. 3%磷酸为流动相B,采用梯度洗脱,流速1. 0mL·min^(-1),柱温:40℃,莫诺苷和马钱苷的检测波长为240nm,丹皮酚的检测波长为270nm。结果莫诺苷在2. 0223~40. 4660μg·mL^(-1)浓度范围内线性关系良好,平均回收率为99. 91%,RSD为1. 26%(n=6);马钱苷在2. 1660~43. 3200μg·mL^(-1)浓度范围内线性关系良好,平均回收率为99. 40%,RSD为0. 84%(n=6);丹皮酚在4. 6154~92. 3080μg·mL^(-1)浓度范围内线性关系良好。平均回收率为99. 63%,RSD为1. 20%(n=6)。结论该法能够同时测定三组分的含量,而且简便、快速、准确,可用于桂附地黄丸(浓缩丸)的质量控制。
OBJECTIVE To establish a method for the determination of total quantity of morroniside and loganin and the content of paeonol in GuifuDihuang Pills (concentrated pills). METHODS High performance liquid chromatography was used to determine the content of morroniside and loganinin FructusCorni and paeonol in peony peel;the chromatographic column was phenomsil C
18 (4.6mm×250mm,5μm),flow phase:with acetonitrile as the mobile phase A,0.3% phosphoric acid as the mobile phase B,the gradient elution,flow rateof 1.0mL·min -1 ,column temperature 40℃,detection wavelength of morroniside and loganin was 240nm,paeonol detection wavelength was 270nm. RESULTS Morroniside in 2.0223~40.4660μg·mL -1 rangelinear relationship is good,the average recovery rate was 99.91%,RSD was 1.26%(n=6);Loganin in 2.1660~43.3200μg·mL -1 range linear relationship
is good,the average recovery rate was 99.40%,RSD was 0.84%(n=6);Paeonol in4.6154~92.3080μg·mL -1 range linear relationship is good,the averagerecovery was 99.63%,RSD was 1.20%(n=6). CONCLUSION The method is simple,rapid and accurate for the simultaneous determination of three components,It can be used for the quality control of Guifu Dihuang Pills (concentrated pills).
