姜黄素对高糖腹膜透析液作用下人腹膜间皮细胞增殖及促纤维化细胞因子表达的影响 被引量:9

摘要 目的:观察姜黄素对高糖腹膜透析液作用下人腹膜间皮细胞(human peritoneal mesothelial cells,HPMCs)增殖及促纤维化细胞生长因子表达的影响。方法:原代培养HPMCs,随机分组为,正常对照组、不同浓度高糖腹膜透析液(PDF)PDF(1. 5%、2. 5%、4. 25%)组和4. 25%PDF+不同浓度姜黄素(20、40、80μmol/L)组,采用CCK-8、real time RT-PCR及ELISA法测定姜黄素对高糖PDF作用下HPMCs增殖及其分泌TGF-β1、CTGF的影响。结果:高糖能显著抑制HPMCs的增殖,上调HPMC[HPMCs]s表达致纤维化细胞生长因TGF-β1及CTGF。不同剂量姜黄素可不同程度抑制高糖PDF引起的上述改变。结论:姜黄素可能通过逆转高糖PDF对HPMCs的增殖抑制作用,下调致纤维化因子TGF-β1及CTGF的表达,进而修复高糖PDF所致的HPMCs的损伤,延缓腹膜纤维化。 Objective:To investigate the effects of Curcumin(Cur) on the inhibition of cell proliferation and expression of transforming growth factor-β1( TGF-β1) and connective tissue growth factor( CTGF) in human peritoneal mesothelial cells(HPMCs) cultured in high glucose-based peritoneal dialysis fluids(PDF). Methods:HPMCs were isolated from human colic omentum by trypsin digestion method and cultured. HPMCs were then divided into normal control group,high glucose group,high glucose-based PDF(1. 5%,2. 5%,4. 25%) group and high glucose-based PDF(4. 25%) plus low-,medium-and high-concentration Cur(20,40,80 μmol/L) group. Counting kit-8( CCK-8) method was used to measure the effects of Cur(20,40,80 umol/L) on cell proliferation. Real time RT-PCR was used to detected the expression of TGF-β1 and CTGF mRNAs in HPMCs. TGF-β1 and CTGF in cultured media were determine by enzyme linked immunosorbent assay( ELISA). Results:High glucose-based PDF significantly inhibited cell proliferation as well as induced the expression of TGF-β1 and CTGF in HPMCs( P〈0. 05). Low-,medium-and high-concentration Cur significantly improved the viability of HPMCs inhibited by high glucose-based PDF. In addition,Cur significantly decreased TGF-β1 and CTGF expression in a dose-dependent manner both at protein and mRNA levels. Conclusion:Curcumin could reverse cell proliferation,and inhibit TGF-β1 and CTGF expression in HPMCs induced by high glucose-based PDF,suggesting that Curcumin can repair the cell damage induced by high glucose-based PDF and delay peritoneal fibrosis.
