脾虚湿阻证动物免疫水平改变及薏苡仁作用机制探讨 被引量:21

Changes of Immune Function of Spleen Deficiency and Dampness Retention(SDDR) Rat Models and The Mechanism of Coix Seed Treatment

摘要 目的:探讨脾虚湿阻证动物免疫水平变化及薏苡仁不同组分健脾利湿作用机制。方法:薏苡仁不同组分干预脾虚湿浊证动物模型,比较其免疫球蛋白IgA、IgG变化情况;并采用基因芯片对其空肠差异基因进行检测,对差异基因进行富集分析,获得Immune Response功能注释到的差异基因后,对相关差异基因进行主成分分析和聚类分析。结果:模型和对照组体质量、白蛋白水平降低且差异具有统计学意义(P〈0.05);多糖、蛋白和油组白蛋白水平有所提高且达到正常对照水平(P〉0.05);模型组IgA、IgG下降,与对照组相比差异具有统计学意义(P〈0.05);各组分治疗后IgA、IgG水平上升,与模型组比较差异具有统计学意义(P〈0.05);对基因芯片进行分析后显示,Immune Response共获得差异基因28个,以累积贡献率〉85%为依据,共获得3个主成分,模型组对照组区分程度良好;各组分治疗后差异基因改善明显,尤其是油、多糖、淀粉和水煎液组,蛋白组对免疫功能调节作用较弱,更接近与模型组;聚类分析也显示,多糖、水煎液、油和正常对照更接近与一类,而蛋白组和模型组更接近与一类;作用的主要基因为Ccl7、Azgp1、Ccr7、Zap70、Tnfsf9、Lat、Lax1、Bpi、Ccl21、Plscr1、Cxcl10、Cxcl9、Tnfsf8。结论:脾虚湿阻证大鼠免疫水平降低,薏苡仁各拆分组分均具有健脾利湿作用,油功效最为显著,其机制与趋化因子和抑癌因子有关。 Objective: To evaluate immune function change of the spleen deficiency and dampness retention(SDDR) rat models and the mechanism of the Coix seed treatment. Methods: The the SDDR rat model was established and treated by the split of the coix seed. The IgA and IgG were compared among different treatment groups. Then the gene microarray change in the jejunum was analyzed. The different expressed genes(DEGs) annotations to the immune response were gained by GO analysis. Then the genes were analyzed by the principal component analysis(PCA) and the enrichment analyses. Results: Model animals had lower IgA and IgG compared with the control group(P〈0.05). After treatment, the IgA and IgG increased obviously compared with those of the model group(P〈0.05). The gene microarray changes showed there was 28 mRNA related to the immune response. Three principal components were gained by PCA. The DEGs improved obviously after treatment by the Coix seed split, especially the oil group, the polysaccharide group,the starch group and the decoction group. On the contrary, the protein group had less effect. The enrichment analysis showed that the DEGs of the polysaccharide group, oil group and the decoction group were near to tha tof the control group, while the DEGs of the protein group and the model group presented the approximate feature. The main DEGs were Ccl7,Azgp1,Ccr7,Zap70,Tnfsf9,Lat,Lax1,Bpi,Ccl21,Plscr1,Cxcl10,Cxcl9,Tnfsf8, etc. Conclusion: SDDR rat possessed of the immunodeficiency. The split of Coix seed can improve the immune function. Especially the oil of the Coix seed has the most prominently effect. The mechanism is relate to the chemokines and the tumor-suppressor factors.
