独活香豆素对乳胞素诱导的帕金森病模型大鼠血清炎症因子的影响 被引量:9

摘要 目的:探讨独活香豆素对帕金森病模型大鼠血清炎症因子的影响。方法:利用乳胞素(Lactacystin)脑黑质致密部注射建立帕金森病(PD)大鼠模型,于第7、14、21、28天进行阿扑吗啡(APO)检测,根据大鼠行为学异常程度,应用分层随机法将模型复制的大鼠随机分为模型对照组、吐温80组、独活香豆素组、独活颗粒剂组、独活复方颗粒剂组、塞来昔布组、美多芭组7组。另设假手术组和正常对照组,假手术组脑黑质致密部注射等量生理盐水。正常对照组、假手术组、模型对照组用生理盐水灌胃,其他组大鼠同时间予以等体积相应药物灌胃,共10周。给药后第14、28、42、56、70天分别进行APO检测,观察记录其旋转行为异常、旋转方向、旋转圈数。在进行最后一次APO检测后的次日,每组随机取8只大鼠釆用ELISA方法检测其血清中IL-1α、IL-1β、IL-6及TNF-α的含量。结果:独活香豆素能改善PD模型大鼠行为学异常,降低血清IL-1α、IL-1β、IL-6、TNF-α炎症因子的异常升高。结论:独活香豆素有可能是通过降低大鼠血清IL-1α、IL-1β、IL-6、TNF-α炎症因子的表达,对PD起到保护作用。 To investigate the effect of Du Huo coumarins on serum inflammatory factors in rats with Parkinson's disease.Method:A rat model of Parkinson's disease(PD) was established by injection of Lactacystin into the Substantia Nigra.APO test was conducted on the 7 th, 14 th, 21 st and 28 th days.According to the degree of behavioral abnormality in rats, the replicated rats were randomly divided into seven groups, namely, the model control group, Tween 80 group, the Du Huo coumarins group, the Du Huo granule group, the Du Huo compound granule group, the Celecoxib group and the Madopar group.The sham operation group and the normal control group were set up separately.In the sham operation group, normal saline was injected into the dense part of the brain substantia nigra.Normal control group, sham operation group and model control group were given gastric lavage with normal saline, while other groups of rats were given gastric lavage with equal volume of corresponding drugs at the same time, a total of 10 weeks.APO test was conducted on the 14 th, 28 th, 42 nd, 56 th and 70 th days after the administration, and the abnormal rotation behavior, rotation direction and rotation number were observed and recorded.On the next day after the last APO test, 8 rats in each group were randomly selected for ELISA to test the serum levels of IL-1α,IL-1β,IL-6,TNF-α.Result:Du Huo coumarins can improve the behavioral abnormality in PD model rats and reduce the abnormal increase of serum IL-1α,IL-1β,IL-6,TNF-α and inflammatory factors.Conclusion:Du Huo coumarins may have a protective effect on PD by decreasing the expression of serum IL-1α,IL-1β,IL-6,TNF-α and inflammatory factors in rats.
