疏肝化瘀方对冠心病心肌缺血大鼠HGF蛋白表达及心功能的影响

Analysis of the Effect of Shugan Huayu Recipe on the Expression of HGF Protein and the Improvement of Cardiac Function in Coronary Heart Disease

目的：探讨疏肝化瘀方对冠心病心肌缺血大鼠HGF蛋白表达及心功能的影响。方法：选取健康雄性SD大鼠72只,采用随机数字表分为假手术组、模型组、阳性对照组、实验组各18只,假手术组和模型组仅给予等量生理盐水处理、阳性对照组给予麝香保心丸治疗、实验组给予疏肝化瘀方治疗;采用免疫组化技术检测各组大鼠干预14 d后的心肌组织中HGF蛋白表达,测定各组SD大鼠心脏功能参数及血清乳酸脱氢酶（LDH）、肌酸激酶（CK）、超氧化物歧化酶（SOD）、一氧化氮（NO）。结果：模型组大鼠心肌组织的HGF蛋白光密度值显著的高于假手术组（P〈0.05）;阳性对照组和实验组大鼠心肌组织的HGF蛋白光密度值显著的高于模型组（P〈0.05）;模型组大鼠LVEDP显著的高于假手术组（P〈0.05）,LVSP、＋dp/dtmax、-dp/dtmax绝对值显著的低于假手术组（P〈0.05）;阳性对照组和实验组大鼠LVEDP显著的低于模型组（P〈0.05）,LVSP、＋dp/dtmax、-dp/dtmax绝对值显著的高于模型组（P〈0.05）;模型组大鼠血清LDH、CK水平显著的高于假手术组（P〈0.05）,SOD、NO水平显著的低于假手术组（P〈0.05）;阳性对照组和实验组大鼠血清LDH、CK水平显著的低于模型组（P〈0.05）,SOD、NO水平显著的高于模型组（P〈0.05）。结论：疏肝化瘀方治疗冠心病心肌缺血大鼠能显著改善其心功能、对心肌具有一定的保护作用,可提高心肌组织HGF蛋白表达水平。

Objective：To explore the effect of Shugan Huayu Recipe the treatment of myocardial ischemia on the expression of hepatocyte growth factor（HGF） protein and cardiac function from the treatment of myocardial ischemia.Methods：Selected 72 healthy male SD rats, were randomly divided into sham operation group, model group, positive control group, 18 rats in each experimental group, sham operation group and model group were given normal saline, the positive control group was given Shexiangbaoxin pill treatment, experimental group received Shugan Huayu Recipe treatment; the expression of HGF protein were detected by immunohistochemical methods in rats after 14 days of intervention in the myocardial tissue, determination of cardiac function parameters and serum LDH group of SD rats（LDH）, creatine kinase（CK）, superoxide dismutase（SOD）, nitric oxide（NO）.Results：The optical density of HGF protein in myocardium of model group was significantly higher than that of sham operation group（P〈0.05）. The optical density of HGF protein in myocardial tissue of model group was significantly higher than that of model group（P〈0.05）. LVEDP in model group were significantly higher than those in sham-operated group（P〈0.05）, LVSP and the absolute volue of ＋ dp/dtmax,-dp/dtmax were significantly lower than those in sham-operated group. The LVEDP in the experimental group were significantly lower than those in the untreated group（P〈0.05）, but LVSP and ＋ dp/dtmax, the absolute value of-dp/dtmax were significantly higher than those in the untreated group（P〈0.05）, and the levels of SOD and NO were significantly lower than those in the sham operation group（P〈0.05）. The levels of serum LDH and CK in the positive control group and the experimental group were significantly lower than those in the model group（P〈0.05）. The levels of SOD and NO were significantly higher than those in the model group（P〈0.05）.Conclusion：Shugan Huayu recipe can significantly improve its cardiac function, have some protective effect on the myocardium and increase the expression of HGF protein in myocardial tissue.
