麝香酮对戊四氮慢性点燃癫痫大鼠的抗痫及脑保护作用研究

摘要 目的:探讨麝香酮对戊四氮慢性点燃癫痫大鼠的抗痫及脑保护作用。方法:将健康雄性sD大鼠90只随机分成6组:空白对照组、模型对照组、麝香酮小剂量组、麝香酮中剂量组、麝香酮大剂量组及丙戊酸钠组,每组各15只。通过行为学观察及脑电图检测,分析探讨不同剂量麝香酮对戊四氮慢性点燃癫痫大鼠首次出现癫痫发作时间、各阶段发作级别、发作潜伏期及持续时间、点燃成功率和癫痫波发放的影响,以探讨其抗痫作用。通过组织形态学检测,探讨麝香酮对脑组织保护作用。结果:麝香酮中剂量及大剂量组首次出现癫痫发作时间均较模型组延迟(P〈20.05,p〈0.01),发作级别降低(P〈o.05,P〈0.01),发作潜伏期延长(P〈0.01),发作持续时间缩短(P〈0.01),癫痫点燃率降低(P〈0.01),癫痫发作波频率和波幅降低。麝香酮大剂量组作用强于中剂量组(1P〈0.05),小剂量组未显示明显抗痫作用。组织形态学结果显示,模型组及小剂量组大鼠脑皮质神经元大量破坏及丢失,而中及大剂量麝香酮组大鼠脑皮质神经元保存相对完整。结论:麝香酮具有一定的抗痫及脑保护作用,其作用强度与剂量呈正相关,相关机制有待进一步研究明确。 Objective: To explore brain protection and antiepileptic effect of muscone in rat kindling models with epilepsy chronically induced by pentylenetetrazol. Methods: 90 healthy male SD rats were randomly divided into 6 groups: the blank control group, the model group, the low-dose muscone group, the middle-close muscone group, the high-close muscone group and the valproate group, with 15 rats in each group. The first seizure time, seizure levels at each stage, the latent period and duration of seizures, the success rate of kindling models with epilepsy and epileptic wave discharge rate were analyzed to explore the antiepileptic effect of muscone in rat kindling models with epilepsy chronically induced by pentylenetetrazol in different dose groups. The histomorphology was used to explore the brain protection effect of muscone. Results:Compared with the model group , the first seizure time in the middle-dose and high- dose groups was later (P〈0.05, P〈0. 01), the seizure levels were lower (P〈0.05, P〈0.01), the latent period of seizures was longer(P〈0.01), the duration of seizures was shorter(P〈0.01), the success rate of kindling models with epilepsy was lower(P〈0.01), and the trequency and amplitude of epi- leptic waves was lower. The antiepileptic effect of high-dose muscone was stronger than that of middle- dose muscone. The low-dose muscone did not show significant antiepileptic effect. The histomorphology results showed that lots of cortical neurons of rats in the model group and low-dose group were destroyed seriously or lost, while the cortical neurons of rats in the middle-dose and high-dose groups were relatively intact. Conclusion: Muscone has some brain protection and antiepileptic effect with posltive dose-effect correlation and the related mechanism needs further study to confirm.
