摘要 目的:检测长链非编码核糖核酸(long non-coding RNA,IncRNA)在癫痫患者外周血液中的异常表达,探究lncRNA能否作为癫痫的生物标记物。方法:采用lncRNA芯片,抽取癫痫患者和正常健康对照者(各5例)的外周血样本,分离出单核细胞(PBMC〉并初步检测表达的lncRNA,寻找两组间表达异常的lncRNA。再根据结果,进一步扩大样本量,选择其中10个显著表达异常的lncRNA在58例癫痫患者和58例正常健康对照者中采用实时荧光定量(qRT-PCR)方法来验证已经筛查出的这10个lncRNA的相对表达水平。结果:病例组血浆NONHSAG012869(PR3)、NONHSAT006265(PR4)、ENST00000581634(PR6)和ENST00000524610(PR8)表达水平分别为2.99±1.59、2.29±0.77、4.24±1.51和11.08±2.24,较对照组的3.74±1.28、2.87±0.90、4.65±0.99、12.00±1.95显著下调(P〈0.05~0.001)。ROC曲线分析显示,4种lncRNA作为联合生物标记物对癫痫具有较高诊断价值(AUC为0.712,敏感度和特异度分别为33.8%和90.6%);Logistic回归分析显示PR8具有最高相对危险度(Odd Ratio为2.459)。结论:PR3、PR4、PR6和PR8可能成为癫痫的联合生物标记物。 Objective:To explore the expression level of long non-coding RNA (lncRNA) in an effort to validate lncRNA as specific epilepsy biomarkers. Methods:The expression of lncRNAs in peripheral blood mononuclear cells (PBMCs) was measured by Agilent Human lncRNA array. 10 lncRNAs were chosen for further validation with quantitative real-time reverse transcription polymerase chain reaction (qRT-PCR) in larger samples including 58 cases and 58 controls, Results:THE expressed levels of NONHSAG012869 (PR3), NONHSAT006265 (PR4), ENST00000581634 (PR6) and ENST00000524610 (PR8) in patients with epilepsy were 2.99±1.59,2.29±0.77, 4. 24±1.51,11.08±2.24 respectively, significantly (P〈0.05 - 0.001), lower than those of the control group(3.74 ± 1.28,2. 87 ± 0. 90,4.65 ± 0. 99 and 12.00±1.95 respectively. ROC curves analysis revealed that the four lncRNAs combined as a panel had significantly diagnostic value in differentiating epilepsy (AUC was 0. 712, sensitivity and specificity were 33.8% and 90.60% respectively); Logistic regression analysis demonstrated that PR7 had the greatest relative risk against epilepsy (Odd Ratio was 2.459). Conclusion:PR3, PR4, PR6 and PR8 may serve as specific biomarkers for diagnosis of epilepsy.
