摘要 目的探讨PI3K抑制剂2-(4-吗啉基)-8-苯基-4氢-1-苯并吡喃-4-酮(LY294002)对人肾透明细胞腺癌786-o细胞PI3K-Akt-m TOR信号通路的影响。方法采用RT-PCR法分别检测LY294002在不同浓度及时间点对786-O细胞处理后细胞的PI3K、m TOR、Akt mRNA的表达情况,探讨其对PI3K、m TOR、Akt mRNA表达的影响。结果在LY294002处理细胞48 h,采用1、2.5、5、10、20、30、40μg/m L浓度处理下,PI3K、Akt、m TOR mRNA的表达显著受到抑制,并且呈现出随药物浓度的变化出现抑制程度增大的正比关系。结论 LY294002可以显著抑制PI3K的水平来影响PI3K-Akt-m TOR信号通路,可为肾细胞癌的分子靶向治疗提供依据。 Objective To investigate the effect of LY294002( PI3K inhibitor) on PI3K-Akt-mTOR signaling pathway in human renal clear cell adenocarcinoma 786-O cells. Methods The expression of PI3K mRNA,mTORmRNA and AktmRNA in 786-O cells treated with LY294002 at different concentrations and time points were detected by RT-PCR. Results The expression of PI3KmRNA,AktmRNA and mTORmRNA was significantly inhibited by LY294002 at the concentration of 1,2. 5,5,10,20,30 and 40μg/m L for 48 h. There was no proportional relation between the concentration of LY294002 and the degree of inhibition.Conclusion LY294002 can significantly inhibit the PI3K level to influence PI3K-Akt-mTOR signaling pathway. This could provides a theoretical basis for the application of LY294002 in molecular targeted therapy of renal cell carcinoma.
