摘要
目的:研究黑米花青苷复方软胶囊中黑米花青苷、西洋参皂苷、灵芝多糖及二苯乙烯苷4种主要成分中单一组分和复配组分的抗氧化活性变化规律。方法:以Vc为阳性对照,采用DPPH法、ABTS法及FRAP法3种方法对单一组分和复配组分的抗氧化活性进行评价,并依据中效原理的联合指数(CI)判断2种组分复配是否具有协同抗氧化作用。结果:单一组分中,黑米花青苷抗氧化活性较好,对DPPH·、ABTS+·的清除率可达80.86%和98%以上,FRAP值为1.125 mmol/L。复配物中,2种组分复配CI值结果显示黑米花青苷分别与西洋参皂苷、灵芝多糖及二苯乙烯苷复配时的CIDPPH和CIABTS值均在0.816~0.930之间,CI值<1,说明有协同增效作用;3种组分复配,黑灵二的抗氧化活性较强,对DPPH·、ABTS+·的清除率可达86.26%和98%以上,FRAP值为0.712 mmol/L;4种组分复配,黑西灵二对DPPH·、ABTS+·的清除率可达81.46%和98%以上,FRAP值为0.617 mmol/L。结论:单一组分和复配组均具有很好的抗氧化活性,该研究对黑米花青苷复方软胶囊产品功效评价,以及相关产品开发具有现实意义。
Objective: To define the change law of antioxidant active of four main components in black rice anthocyanin compound soft capsule: black rice anthocyanin(short for BRA), ginseng saponin(short for GS), Ganoderma polysaccharide(short for GP) and two stilbene glucoside(short for TSG), as well as in different combinations of antioxidant activities. Methods: Used Vc as a positive control, in three methods,including the DPPH method, ABTS method and FRAP method, to systemically evaluate the complex change of antioxidant activities of a single and compound extract. The synergistic antioxidant effects on two ingredients compound were analyzed by median-effect principle combination index(CI). Results: In a single component the black rice anthocyanin has relative stronger antioxidant activity on DPPH· and ABTS+· free radical, DPPH· scavenging rate was 80.86% and ABTS+· scavenging rate was more than98%, the FRAP value was 1.125 mmol/L. In compound extract, the CI value of two ingredients compound group show that black rice anthocyanin and Ganoderma lucidum polysaccharide, ginseng saponin, two stilbene glucoside on antioxidant activity has a synergistic effect, the values of CIDPPH and CIABTS between0.816 and 0.930, CI1. Three kinds of compound group, BRA+GP+TSG combination has the stronger antioxidant activity on DPPH·and ABTS+· free radical, DPPH· scavenging rate was 86.26% and ABTS+·scavenging rate was more than 98%, the FRAP value was 0.712 mmol/L. Combination groups of four main components, DPPH· scavenging rate was 81.46% and ABTS+· scavenging rate was more than 98%,the FRAP value was 0.617 mmol/L. Conclusion: All the a single and compound extract have antioxidant activity, the results for the study of effects of black rice anthocyanin compound soft capsule and the development of related products has the realistic meaning.
