刺梨金银花速溶茶珍的制备及其对CCl_4诱导大鼠急性肝损伤的保护作用 被引量:1

Preparation of Rosa roxburghii and Lonicera japonica Thunb. instant tea and investigation of its protective effect on acute hepatic injury induced by CCl_4 in rat

摘要 目的:确定刺梨金银花速溶茶珍最佳提取工艺,研究刺梨金银花速溶茶珍对CCl_4诱导的大鼠急性肝损伤的保护作用。方法:以总浸膏得率、绿原酸以及总黄酮提取率作为提取工艺的考察指标,采用L_9(3~4)正交试验法,考察加水量、提取时间和提取次数对煎煮工艺的影响;将48只清洁级SD大鼠,随机分为6组(空白对照组、CCl_4肝损伤模型组、联苯双酯阳性药物组、刺梨金银花速溶茶珍高、中、低剂量组),用2%CCl_4花生油溶液制备急性肝损伤大鼠模型,并检测血清中碱性磷酸酶(ALP)、乳酸脱氢酶(LDH)、还原型谷胱甘肽(GSH)、总蛋白(TP)、丙二醛(MDA)、甘油三酯(TG)的含量。结果:最佳工艺为A3B1C2,即加18倍量的水、煎煮2次、每次煎煮1 h;与空白对照组比较,CCl_4肝损伤模型组大鼠血清中ALP和LDH活性明显升高,GSH、TP、MDA、TG的含量明显增加;与CCl_4肝损伤模型组比较,刺梨金银花速溶茶珍高、中、低剂量组均能显著降低CCl_4急性肝损伤大鼠血清中ALP、LDH的活性,降低GSH、TP、MDA、TG的含量。结论:优选得到的刺梨金银花速溶茶珍提取工艺稳定、可行;刺梨金银花速溶茶珍通过降低ALP、LDH活性及GSH、TP、MDA、TG的含量,对CCl_4造成的大鼠急性肝损伤具有显著的保护作用。 Objective: To investigate the optimization process of Rosa roxburghii and Lonicera japonica Thunb. instant tea, and investigate its protective effect on acute hepatic injury induced by CCl4 in rat.Methods: with the dry extract yielding rate and the transport rate of chlorogenic acid and total flavonoids as the examination targets of extraction process, the L9(34) orthogonal design was adopted to optimize the extraction process, and water amount, extraction duration and extraction frequency as testing factors.Rat were randomly divided into six groups, included blank group, model group, bifendate group and high, middle, low does group of Rosa roxburghii and Lonicera japonica Thunb. instant tea. Acute hepatic injury rat model was induced by 0.2% CCl4. Level of alkaline phosphatase(ALP), lactate dehydrogenase(LDH), total protein(TP), malonialdehyde(MDA), triglyceride(TG) in serum were detected. Results:The optimization process of Rosa roxburghii and Lonicera japonica Thunb. instant tea was adding 18-fold of water extracted two times for 1 hour each time. Compared with the blank group, activities of ALP,LDH and level of TP, MDA, TG in model group were significantly increased. Compared with the model group, activities of ALP, LDH and level of TP, MDA, TG in instant tea group were significantly decreased.Conclution: The extraction process of instant tea is stable and feasible. Rosa roxburghii and Lonicera japonica Thunb. instant tea has hepatoprotective effect on acute hepatic injury induced by CCl4 in rat by down-regulation of activities of ALP, LDH and level of TP, MDA, TG.
