摘要
建立高效液相色谱法快速测定保健食品中原花青素含量的方法。样品用甲醇超声提取,将试样溶液和正丁醇:盐酸(95:5,v/v)以及硫酸铁铵溶液置于20 mL顶空瓶中,在100℃烘箱中反应40min后,以甲醇-0.3%磷酸(27:73,v/v)为流动相,经C18色谱柱分离,在525 nm波长处,对保健食品中原花青素含量进行测定。结果表明,该方法在(0.01~0.2)mg/mL范围内线性关系良好,相关系数为0.998,样品的加标回收率为93.4%~98.7%,相对标准偏差小于2%。该方法操作简便、准确可靠、精密度较高,适合于保健食品中原花青素的快速测定。
To establish a rapid method for the determination of procyanidins in health foods by high performance liquid chromatography. The procyanidins of health foods were extracted by methanol, the sample solutions and n-butanol: hydrochloric acid(95:5, v/v) and ammonium ferric ferric sulfate were mixed in 20 mL headspace vial, in heat chambers at 100 ℃ for 40 min. Procyanidins was qualitatively analyzed on a C18 column at 525 nm using methanol-phosphoric acid(27:73, v/v) as the mobile phase. The results showed that an excellent linear relationship in the range of(0.01-0.2)mg/mL(R2=0.998). The recovery ratio of spiked samples from 93.4% to 98.7% with a relative standard deviation less than 2%. The method was simple, accuracy and reliable way to determine the content of procyanidins in health foods.
