摘要
为评估猪睾丸(ST)细胞系作为猪瘟活疫苗生产用细胞基质的可行性,本试验取猪睾丸(ST)基础细胞库、工作细胞库不同代次细胞分别进行致瘤性检验和胞核学检验。结果表明,ST细胞在传代过程中无致瘤性且染色体遗传稳定。本试验取不同代次的ST细胞繁殖猪瘟病毒液并测定其毒液效价;取不同代次的ST细胞繁殖的猪瘟病毒分别对猪进行免疫攻毒测定其免疫原性。结果表明,将ST工作细胞传至第30代,其仍能良好地表达猪瘟病毒,且产毒稳定,病毒滴度高,连续收毒,病毒产量高,其所表达的猪瘟病毒仍保持良好的免疫原性。上述结果表明,猪睾丸(ST)细胞系适宜作为猪瘟活疫苗生产用的细胞基质。
The feasibility of the swine testicle(ST) cell line as a substrate for production of classical swine fever live vaccine has been studied. In this study, the different generation of ST cells of basic and working cell bank was identified by tumorigenic test and karyotype The results showed that the ST cells of working bank passaged 30 times had no difference in karyotype and had no tumorigenicity. The different generations ST ceils were inoculated with classical swine fever virus ( CSFV ) , and the virus title was calculated, and the results showed that the ST cells of working bank passaged 30 times stably produce classical swine fever virus, and the virus title was high. At the same time,the immunogenicity of CSFV from different generations of the ST cells was tested in piglets. The resuits showed that the immunization of CSFV from different generations of the ST cells had good immunogenicity. In conclusion, these results indicate that the ST cells line can be used as a substrate for large scale of production of classical swine fever live vaccine.
