摘要
目的探讨20%瑞香狼毒素对大鼠骨髓和肝脏的遗传毒性效应。方法 SD大鼠50只随机分为5组,20%瑞香狼毒素高、中、低剂量染毒组、阴性对照组和阳性对照组,每组10只,雌雄各半。高、中、低剂量染毒组灌胃量分别为500 mg/(kg.bw)、125 mg/(kg·bw)和31.25 mg/(kg·bw),1次/天,6天/周,连续90 d;阴性对照组给予纯水灌胃对照;阳性对照组按40 mg/(kg·bw)对大鼠进行30 h 2次给药法腹腔注射环磷酰胺。所有动物末次给药后6 h处死。检测各组大鼠胸骨嗜多染红细胞微核率和肝细胞微核率。结果组间比较:20%瑞香狼毒素低、中、高剂量组染毒后骨髓红细胞微核率与阴性对照组比较差异无统计学意义(P>0.05),与阳性对照组比较差异有统计学意义(P<0.05);染毒后,低、中剂量组的肝细胞微核率与阴性对照组比较差异无统计学意义(P> 0.05),高剂量组肝细胞微核率高于阴性对照组(P<0.05)。组内比较:20%瑞香狼毒素高剂量组及阳性对照组的肝细胞微核率高于骨髓红细胞微核率(P<0.05)。结论 20%瑞香狼毒素亚慢性染毒对大鼠骨髓和肝脏微核率的影响不明显,但高剂量染毒时,其对大鼠肝微核率的影响大于骨髓。
Objective To discuss toxicity effect of 20% daphneantoxin on bone marrow and liver of rats.Methods 50 SD rats were randomly divided into 5 groups,with 20% daphrotoxin high-dose group,medium-dose group,low-dose group,negative control group and positive control group. Each group consisted of 10 males and females. For the high-dose group,intragastric intoxication was performed at 500 mg/( kg·bw) for1 time/day,6 days/week,for 90 days in a row; The intragastric volumes of the medium and low-dose groups were 125 mg/( kg·bw) and 31.25 mg/( kg·bw),respectively. In the negative control group,pure water was given; In the positive control group,cyclophosphamide was injected into rats twice for 30 hours with 40 mg/kg. All the animals were sacrificed 6 hours after the last dose. The effects of different doses of20% daphneantoxin on the micronuclear rate of bone marrow and hepatocytes in rats were examined. Results Compared with the negative control group,there was no difference in each dose group( P〉0.05),but different dose groups compared with the positive control group were significant differences( P〈0.05).The micronuclear rate of liver cells in high-dose group was statistically significant compared with other groups( P〈0.05);The high-dose group and the positive control group were statistically significant between bone marrow and liver cell micronuclear rate( P〈0.05). Conclusion The effect of 20% daphneantoxin subchronic poisoning on bone marrow and liver cell micronuclear rate was not obvious,but the effect of high dose on rat liver micronuclear rate was greater than that of bone marrow.
