消退素D1对牙周炎大鼠牙龈组织中IL-1β、IL-6表达的影响 被引量:6

摘要 目的:研究消退素D1(RvD1)对牙周炎大鼠牙龈组织中IL-1β、IL-6炎症因子表达的影响。方法:选取30只雄性SD大鼠,采用双侧上颌第二磨牙绑线法构建大鼠牙周炎模型。建模成功后,将大鼠随机分为5组(1个对照组和4个实验组)(n=6),并采用单次尾静脉注射法给药,4个实验组分别注射1、10、50、100 ng/mL的RvD1,对照组注射等量生理盐水。用药后4周处死所有大鼠,取其双侧上颌第二磨牙周围2 mm的牙龈组织,并制作石蜡切片后,用SP免疫组化染色法检测各组大鼠牙龈组织中IL-1β、IL-6的表达水平。结果:与对照组相比,50、100 ng/mL RvD1组均能明显抑制大鼠牙龈组织中IL-1β、IL-6的表达水平(P <0. 05); 10 ng/mL RvD1组仅明显抑制IL-1β的表达水平(P <0. 05),对IL-6无明显抑制作用(P> 0. 05);最低浓度(1 ng/mL) RvD1对上述两种炎症因子的表达均无影响。不同浓度RvD1对IL-1β的抑制作用依次为100 ng/mL> 50 ng/mL> 10 ng/mL> 1 ng/mL,各浓度组间两两相比均有统计学差异(P <0. 05)。结论:消退素D1可有效抑制牙周炎大鼠牙龈组织中IL-1β、IL-6的表达,且具有浓度依赖性。 AIM: To study the effects of resolvinD1(RvD1)on the expression of IL-1β and IL-6 in gingiva of rats with periodontitis. METHODS: 30 male rats with periodontitis of the maxillary second molar sinduced by ligation method were randomly divided into 5 groups( n =6)and injected with normal saline (the control group) or with RvD1 at 1 ng/mL, 10 ng/mL, 50 ng/mL and 100 ng/mL respectively. 4 weeks after of intervention the rats were sacrificed. The second molars along with surrounding 2 mm gingival tissue were intercepted and prepared for the detection of IL-1β and IL-6 expression in rat gingiva by immunohistochemistry. RESULTS: Compared with the control group, the expression of IL-1β and IL-6 in the gingiva of rats was inhibited by 50 ng/mL and 100 ng/mL of RvD1 ( P 〈0.05 ). 10 ng/mL RvD1 only decreased the expression of IL-1β. 1 ng/mL RvD1 did not affect the expression of the 2 cytokines. 100 ng/mL RvD1 inhibited IL-1β more effective than 50 ng/mL and 10 ng/mL RvD1 ( P 〈0.05). CONCLUSION: RvD1 may inhibite the expression of IL-1β in a concentration-dependent manner.
