摘要
为了确定塔里木马鹿生茸期鹿茸细胞中Ca^2+变化规律,试验以fluo-3/AM荧光指示剂负载采用激光共聚焦技术观察细胞外Ca^2+向胞内流动的情况并测定胞内钙离子浓度,分析鹿茸细胞增殖情况与钙离子浓度的相关关系。结果表明:间充质细胞(MSC)、前成软骨细胞(ACC)易培养,软骨细胞(CC)生长较慢不易培养;CC的Ca^2+浓度较低,补充外源性钙时,CC胞外Ca^2+流入胞内需要的时间较长、需要Ca^2+的量较大。说明鹿茸的生长与MSC、ACC的分化能力有关,鹿茸的钙化与CC的数量、胞外Ca^2+的浓度有关。
In order to determine Variation rule on Ca^2+ in velvet antler cells during antler growth stage in Tarim Red deer,laser confocal microscopy was used via fluo-3/AM as fluorescent indicator to survey the change of Ca^2+flowing status from extracellular to intracellular,and to determine intracellular Ca^2+ concentration,and also to analyze the relationship between the proliferation of velvet antler cells and Ca^2+ concentration. The results showed that mesenchymal cells(MSC)and anterior cartilage cells(ACC)were easy to culture,while cartilage cells(CC)grew slowly and were not easy to culture. The concentration of Ca^2+ in CC was low,and a long time and high amount of Ca^2+ were needed for Ca^2+flowing from extracellular to intracellular when exogenous Ca^2+ was supplemented. The results suggested that the growth of velvet antler had relationship with differentiation ability of MSC and ACC,and the calcification of velvet antler was relative to the amount of CC and the concentration of extracellular Ca^2+.
