摘要
为了调查分析目前牛群中结核病的实际感染状况,试验采用组织病理学筛查,菌株分离,16S rRNA、hsp65和rpoB三基因分析鉴定,抗结核常用药物的药敏测定等方法对某大型屠宰牛场屠宰牛的结核感染状况进行了研究。结果表明:从1 576头屠宰牛中筛选出11个疑似结核菌感染样本,经培养最终获得8株分枝杆菌分离株,分别命名为G46、F90、F20、FJ、Y28D、Y28P、5S、Y5,其中G46、F90、F20、FJ、Y28D、Y28P为偶发分枝杆菌,5S为鸟结核分枝杆菌,Y5为M.conceptionense并且M.conceptionense首次从牛中分离鉴定;16S rRNA鉴定所有菌株均为结核分枝杆菌属,hsp65和rpoB序列分析比对鉴定G46、F90、F20、FJ、Y28D、Y28P与偶发分枝杆菌的相似性为100%,5S和Y5与鸟结核分枝杆菌和M.conceptionense的相似性为100%;除菌株F20外,其余菌株表现出多重耐药性。说明目前牛群中结核病感染状况不容乐观,需要重视。
In order to investigate and analyze the actual situation of tuberculosis infection in cattle herds, histopathological screening, isolation, 16 S rRNA, hsp65 and rpoB gene analysis and drug sensitivity test were used to study the infection status of tuberculosis in cattle slaughtered in a large cattle farm. The results showed that 11 samples were screened from 1 576 slaughtered cattle and 8 Mycobacterium isolates were obtained, named G46, F90, F20, FJ, Y28 D, Y28 P, 5 S, Y5 respectively. Among them, G46, F90, F20, FJ, Y28 D, Y28 P were M. fortuitum, 5 S was M. avium, Y5 was M. conceptionense. M. conceptionense was isolated and identified from cattle for the first time. All strains were identified as Mycobacterium by 16 S rRNA analysis. The similarity between G46, F90, F20, FJ, Y28 D, Y28 P and M. fortuitum was 100%, between 5 S, Y5 and M. avium, M. conceptionense was also 100% by sequence analysis of hsp65 and rpoB. Except for strain F20, the others showed multidrug resistance. The results indicated that the current situation of tuberculosis infection in cattle was not optimistic and more attention should be given to this.
