中药熄风化湿方对腹泻型肠易激综合征大鼠脑肠肽影响的研究

Effect of Xi-Feng-Hua-Shi prescription on brain-gut peptide in the rat with diarrhea-predominant irritable bowel syndrome

[目的]观察熄风化湿方对腹泻型肠易激综合征大鼠血浆和结肠组织5-HT和VIP含量的影响,探讨其对腹泻型肠易激综合征的作用机制。[方法]通过母婴分离+乙酸灌肠法建立IBS-D大鼠模型,将Wistar大鼠随机分为正常对照组、IBS-D模型组、低剂量熄风化湿组、高剂量熄风化湿组和西药组,低剂量熄风化湿组、高剂量熄风化湿组和西药组分别给低剂量熄风化湿方、高剂量熄风化湿方和匹维溴铵混悬液灌胃2周后,检测大鼠稀便率和腹壁撤退反射评分(AWR),ELISA检测血浆5-HT和VIP含量,Western Blot检测结肠组织5-HT和VIP含量。[结果]与IBS-D模型组比较,各给药组大鼠的稀便率、AWR评分、血浆和结肠组织5-HT和VIP含量显著降低,且随着中药浓度的增高,稀便率、AWR评分、血浆和结肠组织5-HT和VIP含量降低更加明显,差异有统计学意义(P<0.05)。[结论]熄风化湿方可能通过降低IBS-D大鼠血浆和结肠组织5-HT和VIP含量来发挥疗效。

[Objective]To observe the effect of Xi-Feng-Hua-Shi prescription on brain-gut peptide in the rat with diarrhea-predominant irritable bowel syndrome（IBS-D）.[Methods]IBS-D model was induced by separation of breast milk＋coloclysis of acetic acid.Wistar rats were divided randomly into control group,model group,low,high dosage Xi-Feng-Hua-Shi group and western medicine group.Low,high dosage XiFeng-Hua-Shi group rats were given by gavage with different concentration XFHS decoction respectively.Western medicine group was given by pinaverium suspl.And equal amount of normal saline was administrated by gavage in the normal group and model group.The abdominal withdrawal reflex was scored,and the level of 5-HT,VIP in the colonic tissue and serum were detected by western blot and ELISA respectively.[Results]Comparing with model group,the grades of AWR and the level of 5-HT,VIP in the colonic tissue and serum in the low,high dosage Xi-Feng-Hua-Shi group and western medicine group were significantly lower（P〈0.05）,with the concentrations of herbs increased,the grades of AWR and the level of 5-HT,VIP in the colonic tissue and serum decreased obviously,and the difference had statistical meaning.[Conclusion]Xi-Feng-Hua-Shi prescription had a therapeutic effect on IBS-D by reducing the level of 5-HT,VIP in the colonic tissue and serum.
