亚硝酰氢对离体大鼠心脏的保护作用

摘要 目的探讨亚硝酰氢对离体大鼠心脏的保护作用。方法取32只Wistar大鼠,麻醉后开胸取出心脏,固定于Langendoff离体心脏灌流装置下端,在恒压条件下灌流离体心脏,选取血流动力学稳定的大鼠心脏18只,随机分为对照组(n=6)、Angeli’s salt(AS)低浓度组(n=6)、AS高浓度组(n=6),分别用KH液、含AS10μmol/L、100μmol/L的KH液灌流,待系统稳定后记录心率(HR)、左心室内压力最大上升速率(+dp/dt)、左心室内压力最大下降速率(-dp/dt)及每分钟从心脏右心耳流出液体量(冠状动脉血流量),最后收集心脏左心室组织,检测肌浆网钙泵(SERCA2a)活性。结果AS高浓度组+dp/dt、-dp/dt较AS低浓度组明显增加,且都高于对照组[(2298.1±216.8)mmHg/s比(1892.6±201.2)mmHg/s比(1545.3±188.6)mmHg/s、(2216.8±254.3)mmHg/s比(1821.7±207.3)mmHg/s比(1482.3±142.9)mmHg/s,P〈0.05];AS低浓度组冠状动脉血流量高于AS高浓度组,且都大于对照组[(8.50±0.86)ml/min比(7.60±50.73)ml/min比(6.80±0.62)ml/min,P〈0.05];AS高浓度组SERCA2a活性较AS低浓度组明显增加,且都高于对照组(8.90±0.90比8.20±0.76比7.40±0.62,P〈0.05);三组间心率比较未见统计学差异[(282.1±14.6)次/min比(279.6±12.6)次/min比(285.2±15.3)次/min,P〉0.05]。结论亚硝酰氢可增加离体心脏冠状动脉血流量,增强SERCA2a蛋白活性,改善离体心脏收缩及舒张功能,对离体大鼠心脏起保护作用。 Objective To explore whether nitroxyl has protective effect on isolated rat heart. Methods 32 Wistar rats were selected, opened chest and took out hearts after anesthesia, fixed them at the bottom of Langendoff isolated heart perfusion device, and maken perfusion of isolated heart under the condition of constant pressure, selected 18 rat hearts with stable hemodynarnics, randomly divided them into control group (n=6) , Angeli's salt (AS) low concentration group (n=6) and AS high concentration group (n=6), make perfusion with KH solution and KH solution containing AS 10 μmol/L and 100 μmol/L respectively, after system was stable, the heart rate (HR), maximum pressure rise rate in left ventricular (+dp/dt), pressure maximum decline rate in left ventricular (-dp/dt)and volume of fluid from right auricle of heart per minute (coronary arterial blood flow ), finally collect heart left ventricular tissue and the activity of SERCA2a were tested and recorded. Results +dp/dt and -dp/dt of AS high concentration group increased significantly compared with AS low concentration group, and both were higher than the control groupC (2298.1±216.8)mm Hg/s vs. ( 1892.6±201.2 )mm Hg/s vs. ( 1545.3±188.6)mm Hg/s, (2216.8±254.3)mm Hg/s vs. (1821.7±207.3)mm Hg/s vs. (1482.3±142.9)mm Hg/s, P〈 0.05 ] ; coronary blood flow of AS low concentration is higher than that of AS high concentration group, and both are greater than the control group [ ( 8.50±0.86 ) ml/min vs. ( 7.60±0.73 ) ml/min vs. ( 6.80±0.62 ) ml/min, P〈0.05 ] ; SERCA2a activity of AS high concentration increases significantly compared with AS low concentration group, and both were higher than the control group (8.90±0.90 vs. 8.20±0.76 vs. 7.40±0.62, P〈0.05 ) ; and there was no significant difference in HR between the three groups [ ( 282.1 ± 14.6 ) times/rain vs. ( 279.6±12.6 ) times/minvs. (285.2±15.3)times/min, P〉0,051.Conclusion Nitroxyl can increase isolated heart coronary blood flow, enhance SERCA2a protein activity and improve isolated heart systolic and diastolic function, having protective effect on isolated rat heart.
