回医烙灸疗法对脊髓损伤大鼠Notch信号通路中Presenilin1、Hes1蛋白表达的影响 被引量:4

摘要 目的回医烙灸疗法对脊髓损伤大鼠Notch信号通路中Presenilin1、Hes1蛋白表达的影响。方法采用Allen’s法制备急性脊髓损伤模型。实验动物分为假手术组(A组)、模型组(B组)、西药组(C组)、2 h烙灸组(D组)、6 h烙灸组(E组),于第1、7、14、28 d取损伤大鼠脊髓组织,免疫组化法检测大鼠脊髓组织中Presenilin1、Hes1蛋白表达。结果 Presenilin1、Hes1免疫组化染色统计结果表明D、E组小于B、C组,且D组表达率最显著减弱,差异有统计学意义(P <0.01)。结论回医烙灸疗法通过抑制Notch信号通路的表达,促进内源性神经干细胞向神经元细胞和少突胶质细胞的分化,以促进其局部神经的修复,且前期烙灸干预的效果好于后期干预。 Objective Effect of Hui medicine Lao moxibustion on expression of Presenilin1 and Hes1 in notch signaling pathway in spinal cord injury rats. Methods Allen's method was used to make the rats' model of acute spinal cord injury. Experimental animals were divided into fake operational group(group A), model group(group B), treatment group by western medicine(group C), treatment group by Hui medicine Lao moxibustion two hours after manufacturing model(group D), treatment group by Hui medicine Lao moxibustion six hours after manufacturing model(group E). On the 1 st, 7 th, 14 th and 28 th day, the injured spinal cord tissue was taken. The expression of presenilin1 and hes1 in the spinal cord rats was detected by immunohistochemistry. Results Presenilin1, Hes1 immunohistochemical staining showed that the D and E groups were less than B and C, and the expression rate of group D was the most significant, and the difference was statistically significant(P〈0.01). Conclusion By inhibiting the expression of Notch signaling pathway, Hui medicine Lao moxibustion therapy promoted the differentiation of endogenous neural stem cells into neuron cells and oligodendrocytes in order to promote the repair of local nerves, and the effect of pre-Lao moxibustion intervention was better than that of later intervention.
