摘要
沉默信息调节因子(SIRT1)是一种依赖烟碱胺腺嘌呤二核苷酸的Ⅲ类组蛋白去乙酰化酶,与酵母菌中发现的沉默信息调节因子2高度同源,其通过对组蛋白以及多种非组蛋白底物的去乙酰化作用调控细胞代谢、衰老、凋亡及氧化应激等众多生理及病理过程。最近研究发现,SIRT1及其下游的调节通路参与心血管疾病的发生发展过程,并起到保护作用。SIRT1在基础医学研究和临床医学应用中具有重要意义,可能成为心血管疾病防治的新策略和潜在靶点。该文对近年来SIRT1在心血管疾病中的研究做一综述。
SIRT1 is a kind of class Ⅲ histone deacetylases which is nicotinamide adenine dinucleotide(NAD + ) -dependent,and it is the closest mammalian homologue of yeast silent information regulator -2 (Sir2).SIRT1 can deacetylate histones and several none-histone transcription factors,and then regulates metabolism,aging,apoptosis,oxidative stress,and other physiological or pathological activities. In recent years,studies have confirmedthat SIRT1 and its downstream pathways play a protective role in the process and development of cardiovasculardiseases. SIRT1 has important significance in basic medical research and clinical medical application. It maybecome a new strategy and potential target for prevention and treatment of cardiovascular diseases. This articlereviewes the current researches of SIRT1 in cardiovascular diseases.
