摘要
目的探讨定位性动脉粥样硬化闭塞症模型制作的新方法,并研究灵芝川芎合剂对下肢动脉粥样硬化闭塞症模型兔的治疗效果。方法将新西兰大白兔随机分成空白组、模型组、治疗组和立普妥组(阳性对照),每组10只。采用高脂喂养与动脉注射聚桂醇泡沫硬化剂相结合的方法,建立下肢定位性动脉粥样硬化闭塞症模型,造模6周后给予相应治疗;治疗组采用灵芝(0. 5 g/kg)及川芎(0. 5 g/kg)合剂煎水灌胃,1次/d,治疗4周;立普妥组给予立普妥0. 2 mg/kg灌胃,1次/d,治疗4周;空白组和模型组给予等量生理盐水灌胃。治疗4周后,对兔下肢进行造影,取血测定血清中总胆固醇(TC)、高密度脂蛋白胆固醇(HDLC)和低密度脂蛋白胆固醇(LDL-C)水平,取术侧髂股动脉进行苏木素伊红(HE)染色和EVG染色。结果与空白组比较,模型组TC和LDL-C水平显著升高(P <0. 05),HDL-C水平显著降低(P <0. 05);与模型组比较,治疗组和立普妥组TC和LDL-C水平显著降低(P <0. 05),HDL-C显著升高(P <0. 05)。病理检查结果显示模型组有显著的动脉粥样硬化特征,且血管内膜弹性纤维显著缺失,而治疗组和立普妥组的动脉粥样硬化特征和膜弹性纤维情况较模型组有明显改善。结论高脂喂养与动脉注射聚桂醇泡沫硬化剂相结合的方法可成功构建下肢动脉粥样硬化闭塞症模型。灵芝川芎合剂对下肢动脉粥样硬化闭塞症模型兔有较好的治疗效果。
Objective To establish a new focal atherosclerosis obliterans model and to study the therapeutic effect of Ganoderma lucidum combined with Chuanxiong on lower extremity atherosclerosis obliterans in rabbits. Methods New Zealand white rabbits were randomly divided into control group( n = 10),model group( n = 10),treatment group( n =10) and Lipitor group( positive control,n = 10). The lower extremity atherosclerosis obliterans model was established by high-fat diet combined with artery injected with lauromacrogol. After 6 weeks of modeling,the rabbits were given with appropriate treatment for 4 weeks. After treatment,iliac artery quantitative angiography was performed,and blood samples were collected to detect the level of total cholesterol( TC),low-density lipoprotein cholestrol( LDL-C) and high-density lipoprotein cholestrol( HDL-C). The histopathologic changes were observed using HE staining and EVG staining.Results Compared with the blank group,the level of TC and LDL-C in the model group were significantly increased( P〈0. 05),and the level of HDL-C was significantly reduced( P〈0. 05). Compared with the model group,the levels of TC and LDL-C in the treatment group and the Lipitor group were significantly reduced( P〈0. 05),while the levels of HDL-C were significantly increased( P〈0. 05). The results of pathological observation showed that the model group had significant atherosclerosis and loss of elastic fibers in endangium,while the changes in the treatment group and the Lipitor group were significantly improved compared with the model group. Conclusion The lower extremity atherosclerosis obliterans model is successfully established by high-fat diet combined with artery injected with lauromacrogol. Ganoderma lucidum combined with chuanxiong has a good effect on treatment of lower extremity atherosclerosis obliterans.
