lncRNA RPSAP16和RPS3AP20在新疆哈萨克族EH患者PBMC中的表达及意义 被引量:1

Expression and clinical significance of lncRNA RPSAP16 and RPS3AP20 in peripheral blood mononuclear cells of patients with essential hypertension in Xinjiang Kazakh nationality

摘要 目的探讨新疆哈萨克族原发性高血压(essential hypertension,EH)患者外周血单个核细胞(peripheral blood mononuclear cells,PBMC)中长链非编码RNA(lncRNA) lncRNA RPSAP16和RPS3AP20的表达及其临床意义。方法采用病例对照研究设计,随机选取哈萨克族EH患者30例(高血压组),选取同期同民族健康体检者30例(对照组),两组患者性别、年龄、吸烟、饮酒、高血压家族史比较差异无统计学意义(P>0. 05)。采集两组研究对象的外周血,运用实时荧光定量PCR (qRT-PCR)方法检测PBMC中lncRNA RPSAP16和lncRNA RPS3AP20表达水平。结果两组在体质指数(BMI)、空腹血糖、三酰甘油(TG)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)等方面比较差异无统计学意义(P> 0. 05)。qRT-PCR结果显示高血压组患者PBMC中的lncRNA RPS3AP20表达水平显著高于对照组(10. 23±2. 94 vs 1. 68±0. 50),差异有统计学意义(P <0. 05)。高血压组患者PBMC中的lncRNA RPSAP16表达水平显著低于对照组(0. 60±0. 24 vs 1. 06±0. 21),差异有统计学意义(P <0. 05)。高血压组PBMC中的lncRNA RPS3AP20水平与收缩压呈正相关(r=0. 64,P <0. 05),与总胆固醇(TC)呈正相关(r=0. 53,P <0. 05),与HDL呈负相关(r=-0. 43,P <0. 05)。高血压组PBMC中的lncRNA RPSAP16水平与TC呈负相关(r=-0. 46,P <0. 05),与TG呈负相关(r=-0. 38,P <0. 05)。lncRNA RPS3AP20表达水平取临界值为4. 47时,其ROC曲线下面积为0. 777,敏感度为80. 0%,特异度为73. 3%。结论 lncRNAs可能与新疆哈萨克族原发性高血压发病存在一定的关联。 Objective To investigate the expression and clinical significance of long chain non coding RNA( lncRNA) lncRNA RPSAP16 and RPS3 AP20 in peripheral blood mononuclear cells( PBMC) of patients with essential hypertension in Xinjiang Kazak population. Methods A case-control study was used. Thirty cases of Kazak essential hypertension patients,who were admitted to the first Affiliated Hospital of Shihezi University from October 2017 to March2018,and 30 healthy cases of the same national for physical examination at the same time were enrolled. The general data and peripheral blood of each subject were collected,and the expression of lncRNA RPSAP16 and lncRNA RPS3 AP20 in PBMC was detected by quantitative real-time PCR( qRT-PCR). Results There was no significant difference in clinical indicators between the two groups. qRT-PCR results showed that the expression level of lncRNA RPS3 AP20 in PBMC of hypertension group was significantly higher than that of control group( 10. 23 ± 2. 94 vs. 1. 68 ± 0. 50,P〈0. 05). The expression level of lncRNA RPSAP16 in PBMC of hypertension group was significantly lower than that of control group( 0. 60 ± 0. 24 vs. 1. 06 ± 0. 21,P〈0. 05). The level of lncRNA RPS3 AP20 in PBMC was positively correlated with systolic blood pressure( r = 0. 64,P〈0. 05) and total cholesterol( r = 0. 53,P〈0. 05),as well as negatively correlated with HDL levels( r = 0. 43,P〈0. 05). The level of lncRNA RPSAP16 in PBMC was negatively correlated with total cholesterol( r = 0. 46,P〈0. 05) and triglyceride( r = 0. 38,P〈0. 05). When the cut-off value of lncRNA RPS3 AP20 was set as 4. 47,the area under the ROC curve was 0. 777,with the sensitivity and specificity of 80. 0% and 73. 3%,respectively. Conclusion The lncRNAs may be associated with the incidence of essential hypertension in Xinjiang Kazakh population.
