摘要
本研究以玉米H99、B73的二倍体和单倍体叶片为材料,采用MSAP技术对其DNA甲基化水平及模式进行研究,分析玉米二倍体和单倍体基因组之间的DNA甲基化水平及其模式变化。结果表明,H99和B73的单倍体甲基化敏感扩增多态性分别为79.90%、83.03%,H99和B73的单倍体甲基化敏感扩增单态性分别为20.10%、17.97%,H99单倍体玉米基因组DNA甲基化水平相比较H99二倍体玉米而言,H99单倍体的总甲基化率降低,全甲基化率降低,半甲基化率升高。B73单倍体玉米基因组DNA甲基化水平相比较B73二倍体玉米而言,B73单倍体的总甲基化率升高,全甲基化率升高,半甲基化率降低。玉米单倍体基因组DNA甲基化模式也发生了改变。本研究初步揭示倍性对于玉米基因组DNA胞嘧啶甲基化的影响。
In this research, the diploid and haploid leaves of maize H99 and B73 were used as materials. The methylation sensitive amplification polymorphism (MSAP) was carried out to study the levels and patterns of DNA methylation. The genomic DNA methylation level and its pattern change of the maize diploid and haploid were analyzed. The results showed that DNA methylation polymorphism of haploid maize H99 and B73 were 79.90% and 83.03%, respectively. Also, the results indicated that DNA methylation monomorphsim of haploid maize H99 and B73 were 20.10% and 17.97%, respectively. Compared with H99 diploid maize, the total methylated rate and full methylated rate of H99 haploid maize genomic DNA methylation levels decreased, but hemimethylated rate increased. Furthermore, compared with B73 diploid maize, the total methylated rate and full methylated rate of B73 haploid maize genomic DNA methylation levels increased, but hemimethylated rate decreased. In addition, maize haploid genomic DNA methylation patterns also changed. This study initially revealed the effects of ploidy on cytosine methylation in maize genomic DNA.
