白藜芦醇对Ni_2O_3诱导NIH/3T3细胞氧化损伤保护作用的实验研究

摘要 目的研究白藜芦醇(resveratrol,3,4’,5-trihydroxystilbene,Res)对三氧化二镍(Ni_2O_3)诱导小鼠胚胎成纤维细胞(NIH/3T3)氧化损伤的保护作用。方法实验分为对照组、三氧化二镍(Ni_2O_3)组以及白藜芦醇干预组。对照组Ni_2O_3浓度为0. 00μg/m L,白藜芦醇浓度为0. 00μmol/L,Ni_2O_3组给予终浓度为200. 00μg/m L的Ni_2O_3混悬液。干预组在Ni_2O_3处理前分别加入终浓度为10. 00、16. 00、20. 00μmol/L的白藜芦醇预处理24h,其他处理与Ni_2O_3组一致。采用CCK-8法检测细胞活性;试剂盒检测细胞内活性氧(ROS)、丙二醛(MDA)和谷胱甘肽过氧化物酶(GSH-Px)含量。结果与对照组相比,Ni_2O_3组细胞活性降低,细胞内GSH-Px含量下降,细胞内ROS及MDA含量增高,差异有统计学意义(P <0. 05);白藜芦醇干预组与Ni_2O_3组比较,随着Res浓度的升高,细胞活性升高,细胞内GSH含量增加,细胞内ROS及MDA含量下降,差异有统计学意义(P <0. 05)。结论白藜芦醇对于Ni_2O_3诱导NIH/3T3的细胞损伤具有保护作用,其机制可能与抑制细胞氧化损伤有关。 Objective The protective effects of resveratrol( resveratrol,3,4,5-trihydroxystilbene,RES) on oxidative damage induced by nickel sesquioxide( Ni2O3) in mouse embryonic fibroblast NIH/3 T3 cells was studied. Methods The experiments were divided into control group,Ni2O3 treated group and resveratrol( RES) intervention group. The concentration of Ni2O3 and the concentration of RES in the control group were both 0. 00 μg/m L. The Ni2O3 treated group was given Ni2O3 suspension with a final concentration of 200 μg/m L. The intervention group was treated with RES solution with a final concentration of 10,16 and 20μmol/L before pretreatment,and 24 h was pretreated. Other treatments were in line with the Ni2O3 treated group. Cell viability was measured by CCK-8 method; kits for measuring reactive oxygen species( ROS) levels and malondialdehyde( MDA) and glutathione peroxidase( GSH-Px) levels in cells. Results Compared with the control group,the cell vitality of the Ni2O3 treated group decreased significantly,the GSH-Px content decreased significantly,the intracellular active oxygen( ROS) level and the MDA increased( P 〈 0. 05); compared with the Ni2O3 treated group,cell viability increased with the increase of RES concentration,the level of GSH-Px increased significantly,and the content of ROS and MDA decreased( P 〈 0. 05) in the group of RES. Conclusion Resveratrol has a protective effect on NIH/3 T3 cell damage induced by Ni2O3,and its mechanism may be related to the reduction of cell oxidative damage.
