舒筋活络药祛风湿药性理论的实验研究

Experimental study on the medical property of Shujinhuoluo anti-rheumatic drugs

目的研究5味舒筋活络药祛风湿的中药药性理论。方法采用维甲酸建立大鼠骨质疏松模型,观测祛风湿中药对骨质疏松大鼠骨密度(BMD)、抗骨折力、骨头灰化质量、骨钙素和血液生化指标Ca、P和ALP水平的影响。采用弗氏完全佐剂(FCA)诱导大鼠建立类风湿性关节炎模型,观察其对佐剂关节炎的影响。结果 5味舒筋活络中药均能改善骨质疏松大鼠的眼角出血、皮毛无光泽、活动量减少、食欲下降、腹泻、下颚和腹部脱毛严重及反应速度降低等症状;增加大鼠骨密度,有3味药达标;显著升高血清磷含量,且作用强度较高,5味药均达标。5味药均具有抑制大鼠致炎侧足肿胀的作用,降低佐剂性关节炎大鼠的关节炎指数,且作用强度均达标++以上;显著提高佐剂性关节炎大鼠血清中抗炎因子IL-2的水平,降低致炎因子TNF-α的水平,5味药均达标。结论舒筋活络药大多入肝、肾经,有的归肝经,味多辛、苦,药性偏温;改善大鼠骨质疏松及抗佐剂性关节炎是祛风湿止痹痛药的药效谱之一。

Objective To study the theory of Chinese medicine property of 5 kinds of Shujinhuoluo （relieving rigidity of muscles and activating collaterals） anti-rheumatics drugs. Methods Rat osteoporosis model was established by retinoic acid,and the effects of anti-rheumatics and analgesia on bone density （BMD）,anti fracture,weight of bone ashing,osteocalcin,blood biochemistry index Ca,P and ALP were investigated.Rheumatoid arthritis rat model was induced by Freund′s complete adjuvant （FCA）,and the anti arthritis effects were investaged. Results All of 5 Shujinhuoluo drugs were able to improve the symptoms including gradually appeared canthus bleeding after osteoporosis modelling,lackluster fur,less activity,decreased appetite,diarrhea,severe depilation of jaw and abdomen as well as low reflex reactions;3 of the drugs were able to increase the bone intensity;5 drugs had the ability of significantly increasing phosphorus in serum with high intensity of action.5 drugs were all effective to inhibit inflammation inducing side foot swelling,decrease the index of FCA rat arthritis with the intensity of over ＋＋;significantly increase the level of anti inflammatory factor IL-2 and decrease the level of inflammatory factor TNF- α .All of the 5 drugs reached treatment standard. Conclusion Most of Shujinhuoluo drugs are related to liver and kidney meridian.Some drugs belong to liver meridian,and were tasted hot and bitter with a tendency to be warm.We ascribed the improvement of osteoporosis and inhibition of FCA arthritis to the Shujinhuoluo theory.
