蒙药朱日很滴丸抗动脉粥样硬化作用的研究

Research on the anti-atherosclerotic effect of Mongolia Medicine Zhurihen Dropping Pills

目的研究蒙药朱日很滴丸抗动脉粥样硬化的作用。方法采用高脂饲料+牛血清白蛋白(250mg·kg^(-1))给予日本大耳白兔进行造模,造模期间测定兔摄食量和体质量的变化。第8周造模成功后,在模型组喂养的基础上,朱日很滴丸组分别灌胃给予朱日很滴丸116,367和1 160mg·kg^(-1),每日1次,连续4周。普罗布考组灌胃给予普罗布考溶液26mg·kg-1。在造模开始至给药过程中的第0,4,8和12周测定各组兔血清中的总胆固醇(CHOL)、低密度脂蛋白(LDL)、三酰甘油(TG)及高密度脂蛋白(HDL)的含量;第12周测定各组兔血清中白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、超氧化物歧化酶(SOD)、丙二醛(MDA)和氧化型低密度脂蛋白(ox-LDL)的含量,取主动脉进行油红O染色和苏木素-伊红(HE)染色,观察主动脉内壁的斑块覆盖率及病理改变。结果与模型组比较,朱日很滴丸组血清中TG、LDL和CHOL的含量降低,HDL的含量升高,差异均无统计学意义(P>0.05);血清中IL-6、TNF-α、MDA和ox-LDL的含量显著降低,SOD的含量显著升高,差异均有统计学意义(P<0.01);主动脉斑块覆盖率降低,差异有统计学意义(P<0.01);血管内皮下见较少吞噬脂质的泡沫细胞和纤维斑块形成。结论蒙药朱日很滴丸具有抗动脉粥样硬化作用。

Objective To research the anti-atherosclerotic effect of Mongolia Medicine Zhurihen Dropping Pills. Methods The rabbit model was established by feeding with high fat feedstuff and bovine serum protein （250 mg·kg -1 ）,meanwhile the changes of food intake and body were measured.The Zhurihen groups were given Zhurihen Dropping Pills （116,367 and 1 160 mg·kg -1 ） from the 8th week for 4 weeks on the basis of the model group,once a day.Probucol group were given probucol 26 mg·kg -1 from the 8th week for 4 weeks on the basis of the model group.The levels of total cholesterol （CHOL）,low density lipoprotein （LDL）,triglyceride （TG） and high density lipoprotein （HDL） in serum at 0,4,8 and 12th week were measured.At 12th week,the levels of interleukin-6 （IL-6）,tumor necrosis factor- α （TNF- α ）,malonedialdehyde （MDA）,oxidized low-density lipoprotein（ox-LDL） and superoxide dismutase （SOD） in serum were measured.The oil red staining method was used to measure the coverage of plaque,and the HE staining method was used to observe the pathology of artery. Results Compared with the model group,the CHOL,LDL,TG and HDL levels of the Zhurihen groups had no significant difference （ P 〉0.05）,but the levels of IL-6,TNF- α ,MDA and ox-LDL in the serum were significantly lower （ P 〈0.01）,the levels of SOD in serum and the coverage of plaque was significantly lower （ P 〈0.01）.HE staining showed less phagocytic lipid cell and fibrous plaque formation in subcutaneous vascular in the Zhurihen groups. Conclusion Mongolia Medicine Zhurihen Dropping Pills have the anti-atherosclerotic effect.
