C57BL/6J老化小鼠GV卵母细胞基因表达模式

摘要 实验采用标准环境下饲养至18月龄的C57BL/6J老化小鼠模型取GV卵子,正常的8周龄大小的雌鼠为对照组,通过单细胞RNA-seq测序来研究老化小鼠和正常小鼠基因表达的差异,经生信分析后发现在细胞凋亡、细胞周期和癌症相关通路存在明显差异,而在生殖相关表达基因上虽然也有差异,但差异表达基因很少不具有统计性。结果说明C57BL/6J老化小鼠模型相对于正常小鼠模型来说在基因表达模式上产生了差异,有157个差异基因,差异主要集中在细胞代谢、凋亡和一些老化疾病通路上,而生殖相关表达基因虽然也存在差异,但是不能说明卵母细胞影响了生殖系统基因的表达。 The C57BL/6J aging mouse model used in the experiment was raised to 18 months old under standard conditions, normal 8 weeks old mouse as control group, comparison of gene expression by bioinformation tecnology in aging mouse models and normal control mouse models by single-cell RNA-seq sequencing, the results showed that there are difference in biocarta, cell cycle and cancer pathways,although there are differences in gene expression related to reproduction,few differentially expressed genes are not statistically significant. The conclusion is that the C57BL/6J aging mouse model produced differences in gene expression patterns compared to normal mouse models, there are 157 differential genes and the differences are mainly in cell metabolism, apoptosis, and some aging disease pathways, it does not mean that oocytes affect the expression of genes in the reproductive system.
