酒精对人牙周膜细胞增殖能力影响的体外研究

Effects of Alcohol on Proliferation of Human Periodontal Ligament Cells in Vitro

目的:研究酒精对人牙周膜细胞增殖活性的影响。方法:酶消化法结合组织块法分离培养人牙周膜细胞,取4～6代细胞,在不含酒精培养基中(对照组)及0. 05mol/L、0. 10mol/L、0. 20mol/L不同浓度酒精环境下(实验组)培养6、12、24、48h,使用MTT法测定酒精对人牙周膜细胞增殖的影响。结果:0. 05mol/L、0. 10mol/L、0. 20mol/L浓度实验组人牙周膜细胞增殖能力均低于对照组。0. 20mol/L浓度酒精在24、48h显著抑制人牙周膜细胞增殖,差异有统计学意义(P <0. 05)。结论:不同浓度的酒精抑制人牙周膜细胞增殖,在牙周炎骨吸收中可能起促进作用。

Objective：To explore effect of alcohol on proliferation of human periodontal ligament cells. Methods：The primary hPDLCs were cultivated with enzyme digestion assay and tissue cultivation. The 4 - 6 generation of hPDLCs were respectively cultured 6,12,24,48 hours in 0mol/L（ control group） ,0.05mol/L,0.10mol/L,0.20mol/L concentration of alcohol（ experimental groups）. The proliferation of hPDLCs was assayed by MTT method at 6,12,24,48hours. Results ： The alcohol of 0. 05mol/L,0.10mol/L,0.20mol/L inhabited the proliferation of hPDLCs. The alcohol of 0.20mol/L significantly inhabited the proliferation of hPDLCs at 24,48hours, the difference was significant （ P 〈 0.05 ）. Conclusion ： The alcohol with different concentration can inhibit the proliferation of hPDLCs and may promote bone resorption in periodontitis.
