摘要
目的:建立夜香牛颗粒中绿原酸和木犀草苷含量分析的方法。方法:采用FORTIS-C18柱(4.6 mm×250 mm,5μm),以乙腈-0.05%KH2PO4溶液为流动相,采用梯度洗脱,流速1.0mL·min^(-1),柱温30℃,检测波长330 nm。结果:绿原酸在4.82~96.30μg·mL^(-1)、木犀草苷在1.62~32.40μg·mL^(-1)范围内有良好线性关系(r=0.999 9),平均加样回收率分别为98.8%和98.4%,RSD分别为1.07%和1.64%。结论:本方法灵敏度高、准确、重复性好、专属性强,可用于夜香牛颗粒的质量控制。
OBJECTIVE To establish a method for determination of chlorogenic acid and luteoloside in Vernonia cinerea granules.METHODS The separation was performed on the Fortis-C(18) column(4.6 mm×250 mm,5μm)with acetonitrile and0.05% KH2PO4 as the mobile phase at the flow rate of 1.0 mL·min-1.The column temperature was 30℃,and the UV detector was set at 330 nm.RESULTS There was good linearity between the peak area and concentration in the ranges of 4.82-96.30μg·mL-1 and 1.62-32.40μg·mL-1(r=0.999 9)for chlorogenic acid and luteoloside,respectively.The average recovery rates were 98.8% and 98.4%.RSD were 1.07% and 1.64%.CONCLUSION The method is highly sensitive,accurate,repeatable and specific,so it is an accurate method for Vernonia cinerea granule quality control.
