骨髓嵌合促进Lewis肺腺癌细胞体内生长及转移的实验研究 被引量:1

摘要 目的研究骨髓移植对近交系C57BL/6(C57) 6~8周龄的野生雌性小鼠皮下接种Lewis肺腺癌细胞成瘤及转移能力的影响。方法采用随机数字表法抽取20只野生小鼠作为受体小鼠,通过8 Gy全身照射破坏受体小鼠骨髓造血系统,再通过尾静脉注射向受体小鼠移植正常小鼠的骨髓细胞,建立骨髓嵌合小鼠模型。随后将其分为对照B组和荷瘤组(每组10只),再使用随机数字表法取20只同批次未处理的野生小鼠分为对照A组和荷瘤组(每组10只)。分别向骨髓嵌合小鼠和野生小鼠背部皮下注射Lewis肺腺癌细胞获得皮下荷瘤模型,通过比较起瘤时间,肿瘤增长速度,离体瘤质量及肝、肺的转移情况,评价骨髓移植对C57BL/6小鼠皮下荷瘤模型的影响。结果皮下荷瘤后,骨髓嵌合小鼠较正常小鼠起瘤时间提早2 d,接种后第5天即可触及肿瘤;相同荷瘤时间,骨髓嵌合小鼠肿瘤体积增长更快,且离体瘤质量增加49. 27%(P <0. 01),并可观察到肺部有明显的肿瘤细胞转移。结论骨髓嵌合后C57小鼠相对正常C57小鼠更适于皮下荷瘤造模,其肿瘤生长更大,更容易发生转移。 Objective To investigate the effect of bone marrow transplantation on the tumorigenesis and metastasis of subcutaneously inoculated Lewis lung cancer( LLC) cells in C57 BL/6( C57) mice.Methods After irradiated with 8 Gy carbon-ion beam to destroy the bone marrow hematopoietic system,the mice were transplanted with the bone marrow cells collected from normal C57 mice via tail vein injection to establish a mouse model of bone marrow transplantation( BMT). Then,both BMT mice and normal C57 mice were subcutaneously injected with LLC cells on the back to build a model of lung tumor-bearing mice,Then the tumor size,tumor weight and lung metastasis were recorded to evaluate the effect of BMT. Results The tumor masses could be touched on day 5 after subcutaneous tumor implantation( day 0),which was in 2 d earlier than in the C57 mice. At the time of sacrifice day( day 19),the BMT mice had significantly larger tumor size and heavier weight compared with normal C57 mice( P〈0. 01). Moreover,obvious lung metastasis foci were observed in BMT mice but not in normal C57 mice. Conclusion BMT C57 mice are more suitable for subcutaneous tumor modeling than normal C57 ones,with larger tumor size and easier for metastasis.
