摘要
目的优化仙茅中有效成分仙茅苷的提取工艺,建立仙茅苷含量检测的HPLC方法。方法系统考察了各单因素对仙茅苷提取率的影响,采用正交设计优化了提取工艺,建立了高效液相色谱法(HPLC)测定仙茅提取液中仙茅苷含量的方法。结果最佳的提取工艺为:80%体积分数的甲醇回流提取3 h,料液比为1∶50,此时仙茅苷的提取率为0.244%;建立的HPLC方法精密度、稳定性、重复性和加样回收率良好,灵敏度较高。结论采用正交试验设计得到的仙茅苷提取工艺,具有较高的仙茅苷提取率。该提取工艺及HPLC分析方法可为中药仙茅的质量控制(包括提取及分析)提供可供参考的实验数据。
Objective To optimize the extract rate of Curculigoside from Curculigo orchioides Gaertn.and establish an HPLC method for the detection of Curculigoside.Methods A systemic study about different single factor was conducted to obtain the best extraction process.On the basis of one-factor-at-a-time method,orthogonal experimental design was investigated for further optimize the process.In addition,the chromatographic separation was was also conducted.Results The optimal extraction was obtained as follows:methanol concentration 80% as the extraction solvent of reflux,3 h,solid-liquid ration 1:50(g/ml),and the extraction rates of Curculigoside reached 0.244%.The method developed for detection had the advantage of good precision,stability,repeatability,and a high recovery.Conclusion The best extraction conditions can be obtained with the help of orthogonal experiment,and the extraction rate obtained is satisfying.The extraction conditions and HPLC method developed can provide experimental data for the quality control of Curculigo orchioides Gaertn.
