摘要
目的观察新葛根芩连汤对糖尿病脑病大鼠海马血脑屏障功能及occludin/Claudins-5蛋白表达的影响。方法以高脂饮食加链脲佐菌素腹腔注射复制糖尿病脑病模型,以新葛根芩连汤干预,8周后髂静脉注射Evans Blue观察血管神经屏障渗透状况,并检测紧密连接蛋白occludin/Claudins-5。结果模型组大鼠海马区Evans Blue外渗明显,药物治疗后中剂量组Evans Blue含量明显减低。模型组紧密连接蛋白occludin/Claudins-5表达均明显降低,治疗后表达明显增强。结论新葛根芩连汤可保护糖尿病脑病大鼠海马区血脑屏障,减少血脑屏障损害,机理可能与保护紧密连接蛋白occludin/Claudins-5有关。
Objective To observe the effects of modified Gegenqinlian decoction on hippocampal blood-brain barrier function of diabetic encephalopathy rats and expression of occludin/Claudins-5 protein.Methods Diabetic encephalopathy models were replicated by high fat diet and intraperitoneal injection of SZT,while the treatment groups were respectively given modified GegenQinlian decoction,cilostazol through intragastric.After 8 weeks,the Evans Blue was injected from iliac vein to observe the permeability of blood brain barrier.Meanwhile,the tight junction protein occludin/Claudins-5 were detected.Results The Blue Evans in hippocampus of model group rats exosmosed obviously,however the middle dose group were significantly decreased after treatment.The expression of tight junction protein occludin/Claudins-5 were significantly decreased in model group,and the expression was significantly increased in treatment groups.Conclusion The modified Gegenqinlian decoction can protect diabetic encephalopathy rat blood brain barrier,which is probably related to improve the expression of tight junction proteins,such as occludin/claudins-5.
