摘要
目的探讨血府逐瘀汤对冠心病血瘀证模型大鼠的血浆接触激活系统蛋白(FXII、HK、KLK)含量以及心功能等的影响。方法 32只SD大鼠随机平分为假手术组、模型对照组、血府逐瘀汤高、低剂量组,后3组均结扎冠状动脉左前降支建立冠心病血瘀证模型,灌胃给药1个月后检测大鼠射血分数(EF%),制备血浆用ELISA法测定FXII、HK、KLK含量,取心脏行HE及MASSON染色。结果与模型组比较,血府逐瘀汤高、低剂量两组射血分数均大于模型组,差异具有统计学意义(P <0.05)。四组大鼠FXII的水平差异无统计学意义(P> 0.05)。血府逐瘀汤高、低剂量组HK及KLK水平均明显高于模型组,差异具有统计学意义(P <0.01)。结论血府逐瘀汤可以改善冠心病血瘀证大鼠的心功能及心肌纤维化,同时可以影响接触系统关键蛋白HK、KLK的血浆水平。
Objective To investigate the effect of Xuefuzhuyu Decoction on plasma contact activation system protein(FXII, HK, KLK) and heart function in rats with coronary heart disease with blood stasis syndrome. Methods Thirty-two male SD rats are randomly divided into sham operation group, model control group and Xuefuzhuyu Decoction high-dose group and low-dose group. Except for sham operation group, the left anterior descending coronary artery is ligated to establish coronary heart disease. Blood stasis syndrome model is administered by intragastric administration. One month later, the ejection fraction(EF %) of rats is measured with an ultrasound system. The rats are sacrificed and blood is taken for preparation of plasma. ELISA is used to determine the levels of FXII, HK and KLK, and the heart is taken for sectioning. HE and MASSON staining. Results Compared with the model group, the ejection fraction of the high and low doses of Xuefuzhuyu Decoction is higher than that of the model group, and the difference is statistically significant(P〈0.05). There is no significant difference in the level of FXII between the four groups(P〉0.05). The levels of HK and KLK in the high-dose and low-dose groups of Xuefuzhuyu Decoction are significantly higher than those in the model group, and the difference is statistically significant(P〈0.01). Conclusion Xuefuzhuyu Decoction can improve the heart function and myocardial fibrosis in rats with coronary heart disease and blood stasis syndrome, and can also affect the plasma levels of key proteins HK and KLK in the exposure system.
