小金丹抑制乳腺癌细胞转移及其机制研究 被引量:11

Inhibitory effect of Xiaojindan on the metastasis of breast cancer cells and its related mechanisms

摘要 目的:研究小金丹对乳腺癌细胞侵袭和转移的影响及其作用机制。方法:选择高转移性乳腺癌细胞株MDA-MB-453细胞株,用不同浓度的小金丹提取物(25、50、100、150、200mg生药/L)处理细胞24h后,分别采用肿瘤细胞黏附实验、划痕实验和Transwell侵袭实验检测细胞黏附、运动、侵袭能力;Western blot法检测上皮-间质转化(EMT)相关蛋白和丝裂原活化蛋白激酶(MAPK)通路蛋白的表达水平。结果:与对照组比较,小金丹提取物作用24h后能明显抑制人乳腺癌MDA-MB-453细胞的运动和侵袭能力(P<0.01),作用呈现剂量依赖性关系(P<0.01);与对照组比较,100、150mg/L小金丹提取物能明显增加ZO-1和β-catenin表达(P<0.01),降低Vimentin表达(P<0.01),明显抑制p-JNK和p-p38的蛋白表达水平(P<0.01)。结论:小金丹能有效地抑制高转移性乳腺癌细胞的运动和侵袭能力,作用机制可能与其调控p38 MAPK和JNK MAPK通路,逆转肿瘤细胞EMT有关。 Objective: To study the effect of Xiaojindan(XJD) on invasion and metastasis of breast cancer cells and its mechanism. Methods: High-metastatic breast cancer cell line MDA-MB-453 was selected. Cells were treated with different concentrations of XJD extract(25, 50, 100, 150, 200 mg crude drug/L) for 24 h, respectively. The cell adhesion, movement and invasion ability were detected by Tumor cell adhesionassay, scratchassay and Transwell invasion assay. The expression levels of epithelial-mesenchymal transition(EMT)-associated protein and mitogen-activated protein kinase(MAPK) pathway protein were detected by Western blot. Results: Compared with the control group, XJD could significantly inhibit the movement and invasion of breast cancer MDA-MB-453 cells in a dose-dependent manner after 24 hours(P0.01). Compared with the control group, XJD extract 100, 150 mg/L could increase the expression of ZO-1 and β-catenin(P0.01), and the expression of Vimentin was decreased(P0.01), significantly inhibit the protein expression levels of p-JNK and p-p38(P0.01). Conclusion: XJD can effectively inhibit the migration and invasion of high metastatic breast cancer cells. The mechanism may be related to the regulation of p38 MAPK and JNK MAPK pathway and the reversal of tumor cell EMT.
