菊苣降尿酸药效验证 被引量:29

Pharmacodynamics authentication research on uric acid-lowering active ingredients of Cichorium intybus L.

摘要 目的:初步阐明菊苣降尿酸的物质基础,并对其降尿酸机制、安全性进行初探。方法:迪法克鹌鹑60只(雄性),按体质量随机分为6组,即正常组、模型组、苯溴马隆组(20mg·kg^(-1)·d^(-1))、绿原酸组(150mg·kg^(-1)·d^(-1))、菊苣酸组(150mg·kg^(-1)·d^(-1))、秦皮甲素组(150mg·kg^(-1)·d^(-1)),每组10只。除正常组给予鹌鹑普通饲料外,其余各组均给予高嘌呤饲料(普通饲料拌入酵母料15g·kg^(-1)·d^(-1))复制高尿酸血症模型。给药3周,动态观察鹌鹑尿酸(UA)水平,肝功能指标谷草转氨酶(GOT)、谷丙转氨酶(GPT)水平,肾功能指标肌酐(Cr)、尿素氮(BUN)水平及UA代谢相关酶黄嘌呤氧化酶(XOD)、腺苷脱氨酶(ADA)活性的变化。结果:与正常组比较,第7、14、21天模型组鹌鹑血清UA水平显著升高(P<0.05);与模型组比较,第7、14、21天绿原酸组、秦皮甲素组鹌鹑血清UA水平显著降低(P<0.05),第14、21天菊苣酸组鹌鹑血清UA水平显著降低(P<0.05),第14天秦皮甲素组、菊苣酸组鹌鹑血清GPT水平显著降低(P<0.05),第7、14、21天绿原酸组、菊苣酸组鹌鹑血清XOD水平显著降低(P<0.05),第21天秦皮甲素组、菊苣酸组鹌鹑血清ADA水平显著降低(P<0.05)。结论:菊苣发挥降尿酸药效的物质基础可能为绿原酸、秦皮甲素、菊苣酸。 Objective: To preliminary clarify the material basis of uric acid reduction by Cichorium intybus L., and preliminary study its mechanism and safety of uric acid reduction. Methods: Sixty quails (males) were randomly divided into 6 groups according to their body weight; the normal group, the model group, the benzbromarone group (20mg · kg^-1· d^-1), and the chlorogenic acid group (150rag1 kg^-1· fit), cichoric acid group (150mg · kg^-1· d^-1), aesculin group (150mg · kg^-1· d^-1), 10 in each group. Except for the normal group, the other groups were given high purine feed (the common feed was mixed with yeast material 15g ·kg^-1 ·fit) to replicate the hyperuricemia model. After 3 weeks of administration, the uric acid (UA) level, liver function index GOT, GPT level, renal function index Cr, BUN level and UA metabolism related enzyme XOD, ADA activity were observed dynamically. Results: During the modeling period, compared with the normal group, the UA level of the sputum in the model group was significantly increased in day 7, 14, 21 (P〈0.05). Compared with the model group, the serum UA level of the chlorogenic acid group and the aesculin group was significantly decreased in day 7, 14, 21 (P〈0.05), and the UA level in the chicoric acid group was significantly decreased in day 14, 21 (P〈0.05), the serum GPT level in aesculin group and chicory acid group was significantly decreased in day 14 (P〈0.05), the serum XOD level in cblorogenic acid group and chicory acid group was significantly decreased in day 7, 14, 21 (P〈0.05), the serum ADA level in aesculin group and chicory acid group was significantly decreased in day 21 (P〈0.05). Conclusion: The material basis of Cichorium intybus L. to exert uric acid^-1owering effect may be chlorogenic acid, aesculin and cichoric acid.
