肺岩宁联合吉非替尼对人非小细胞肺癌H1975细胞生物学行为的影响 被引量:2

Influence of Feiyanning with gefitinib on biological behavior of human non-small cell lung cancer cell line(H1975)

摘要 目的:观察比较肺岩宁,吉非替尼及两药联合对肺癌细胞H1975增殖、周期和凋亡等细胞生物学功能的影响。方法:采用MTT法检测肺岩宁(2.5、5、10、20、40mg/mL)、吉非替尼(1、3、5、10、20μmol/L)的最佳作用时间及单药和两药联合干预对H1975细胞增殖的抑制作用;流式细胞术观察肺岩宁(5mg/mL)、吉非替尼(1μmol/L)及联合用药对H1975细胞周期和凋亡的影响。结果:MTT结果显示,肺岩宁和吉非替尼可抑制H1975细胞的生长,两种药物的最佳作用时间均为48h,联合用药组对肿瘤细胞的生长抑制明显强于肺岩宁组和吉非替尼组(P<0.01)。流式细胞结果显示,联合用药组肿瘤细胞凋亡率为(36.12±1.17)%,明显高于肺岩宁组(21.60±1.25)%和吉非替尼组(15.82±2.49)%(P<0.01);且各给组均可不同程度上将细胞周期阻滞在S期。结论:肺岩宁方可以显著抑制人非小细胞肺癌H1975细胞的生长,诱导其凋亡,阻滞细胞于S期;与吉非替尼联合应用可明显起到增效作用。 Objective: To study the effects of Feiyanning(FYN), gefitinib and their combination on the cell biology function of human non-small cell lung cancer(NSCLC) cell line H1975, such asproliferation, cycle and apoptosis. Methods: MTT method was used to detect the optimal concentration and action time of FYN(2.5-40 mg/mL), gefitinib(1-20μmol/L) and the survival of tumor cells treated with single drug and their combination effect. The apoptoticrate and grow thcycle of the cells were detected by flow cytometry. Results: MTT assay showed that FYN and gefitinib could inhibit the growth of H1975 cell line, the optimum reaction time was 48 hours for both drugs. And the inhibition of united group was significantly stronger than that of the FYN group and gefitinib group. Flow cytometry analysis showed that the ratio of apoptosis was(36.12±1.17)% at 48 h for united group, which was significantly higher than the FYN group(21.60±1.25)% and gefitinib group(15.82±2.49)%, the difference was significant(P0.01). The cells in different groups of drugs were mainly blocked in Sphase in varying degrees. Conclusion: FYN can effectively destroy the cell survival and development of tumor, induce apoptosis degradation and blocked the cell cycle in S phase. And there is synergism when combined with gefitinib.
