大足黑山羊妊娠不同阶段胎盘线粒体相关基因的表达 被引量:3

Mitochondrial related genes expression in placental tissues of different gestational stages in Dazu black goat

摘要 胎盘在母体-胎儿间物质交换时发挥着重要作用,而其物质交换动力来自胎盘线粒体通过氧化磷酸化产生ATP。为探讨随着动物妊娠阶段的持续,线粒体相关基因的表达量变化,本研究采用RT-PCR技术检测大足黑山羊妊娠20,25,30,60,90和120d胎盘线粒体COXIV、SDHA、PGC-1α、NRF-1和TFAM基因的相对表达量。结果显示,随着大足黑山羊妊娠持续,胎盘子叶和子宫阜线粒体被检测的5项基因相对表达量均呈现先升高后降低、再缓慢变化的趋势。尤其在妊娠25d时相对表达量均达到高峰,但妊娠30d时相对表达量急剧下降。在胎盘子叶组织中COXIV、SDHA和NRF-1的相对表达量分别由0.84,1.53和1.06,降至0.09,0.32和0.22,差异显著(P〈0.05);在子宫阜中COXIV、SDHA和PGC-1α的相对表达量分别由0.50,4.36和1.36降至0.11,0.84和0.48,差异极显著(P〈0.01)。妊娠60d后,在胎盘子叶组织中,这些基因相对表达量不断升高;在子宫阜,相对表达量呈递减趋势,但差异均不显著(P〉0.05)。相关性分析结果显示,子宫阜组织中线粒体相关基因的表达与胎儿及胎盘生长发育指标如胎儿重、胎儿长、胎盘重、胎盘效率、子叶个数和子叶直径呈显著负相关(P〈0.05);而子叶组织中线粒体相关基因的表达与胎儿长和子叶直径显著性相关(P〈0.05),但与胎儿重、胎盘质量、胎盘效率和子叶个数无显著相关性(P〉0.05)。 Placenta plays an important role in material exchange between maternal and fetal, and the material exchange power comes from production ATP through oxidative phosphorylation in mito- chondria. To investigate the variation of mitochondrial related gene expression with the extension of animal pregnancy stages,RT-PCR was used to detect the relative expression of placental mito- chondrial genes PGC-lα, NRF-1, SDHA, TFAM and COXIV, on the 20,25,30,60,90 and 120 days of pregnancy in Dazu black goat. The results showed that the relative expression levels of the placental cotyledon and uterine caruncle mitochondrial gene, such as COXIV, SDHA, PGC-lα, NRF-1 and TFAM, increased firstly, and decreased subsequently, and then changed slowly with the gestation of Dazu black goat. Especially, it reached a peak at 25 days of pregnancy, but decreased sharply at 30 days of pregnancy. Particularly,the expression of mitochondrial COXIV, SDHA and NRF-1 gene decreased from 0. 84 ,1. 53 ,1. 06 to 0. 09,0. 32,0. 22 in cotyledons,respec- tively,which had significant differences (P〈0.05);and the relative expression of mitochondrial COXIV, SDHA and PGC-1α were reduced from 0. 5,4. 36,1. 36 to 0.11,0. 84,0. 48 in uterine car- uncle,respectively,which had extremely significant differences (P〈0.01). After 60 days of preg- nancy,the relative expression of mitochondrial genes showed ineresed an tend in cotyledon ; and it decreased in uterine caruncle,but the difference was not significant (P〉0.05). Correlation analy- sis showed that the there was a negative relation between the mitochondrial genes expression in u- terine caruncle and the changes in fetal quality,fetal length,placental weight,placental efficiency, cotyledon number and cotyledon diameter (P〉0.05);and there was a significantly relation be tween the mitochondrial related genes expression in cotyledon and fetal length and cotyledon diam- eter (P〈0.05),but there was no si cental efficiency and cotyledon numb gnificant correlation with fetal quality, placental weight, pla er (P〉0.05).
