摘要
KiSS-1基因在雌性动物初情期启动中扮演着重要作用,其启动子区甲基化状态与KiSS-1mRNA表达量之间关系尚不清楚。本试验利用亚硫酸氢盐测序(bisulfite sequencing PCR,BSP)技术和荧光定量PCR技术研究了小尾寒羊初情期前、临近初情期和初情期母羊下丘脑KiSS-1基因启动子区甲基化状态和KiSS-1基因mRNA表达量,分析两者之间的关系。结果表明:随着初情期的启动,KiSS-1基因启动子区甲基化水平显著降低,尤其是在启动子区-501位点降低最明显,并且KiSS-1基因mRNA表达量随着初情期的启动显著升高。结果提示:初情期的启动可能通过降低下丘脑KiSS-1基因启动子区特定位点甲基化水平,从而提高KiSS-1基因mRNA表达量。
The KISS-1 ever, the relationship gene plays an important role in the onset of puberty in female animals. How- between the methylation status of KISS-1 promoter and the expression of KISS-1 mRNA during puberty onset is unclear. In this study, the methylation status of KISS-1 gene promoter and the expression of KISS-1 mRNA in hypothalamus of ewes in different stages (peri-puberty,pre-puberty and puberty)of small tail han sheep was research by using bisulfite se- quencing PCR(BSP)and reahime fluorescence quantitative PCR. The results showed that with the onset of puberty, the methylation level of KISS-1 gene promoter region significantly decreased, es- pecially in the promoter region-501, while the expression of KISS-1 mRNA increased significantly. The studies indicated that the increasing expression of KISS-1 mRNA may be cuased by decreasing the methylation level of the specific site in the promoter region of KISS-1 gene in hypothalamus during the onset of puberty in sheep.
