摘要
目的运用基于GC/MS的血浆代谢组学研究新疆哈族肥胖儿童菌群人源化小鼠特征代谢物,探讨新疆哈族肥胖儿童的可能机制。方法收集新疆哈族肥胖儿童粪便,制备粪便悬液,制备肠道菌群人源化小鼠模型组(HUM组),设置健康小鼠作为对照组(GF组),运用GC/MS对两组小鼠的血浆代谢物进行研究,运用PCA和PLS-DA统计学方法寻找出特征代谢物,鉴别特征代谢物。结果最终获得183个变量。PCA、PLS-DA和VIP分析最终得到21个特征代谢物;结构鉴别发现,HUM组的三羧酸循环中间物柠檬酸、琥珀酸、苹果酸等显著高于GF组;HUM组6磷酸葡萄糖、6磷酸果糖、丙酮酸和草酰乙酸表达水平显著高于GF组。此外,本研究还发现HUM组的马尿酸、如哌啶甲酸、5-氨基戊酸等显著高于对照组。结论肠道菌群可能通过影响哈族儿童体内的三羧酸循环和糖酵解活跃状态,同时肠道菌群代谢物活性增强,促进肠道营养吸收,导致哈族儿童肥胖。
Objective Using GC/MS-based plasma metabonomics to study the characteristics of humanized mouse metabolites in the obese children of Xinjiang Kazakh ethnic group,and to explore the possible mechanism of Xinjiang obese children in Xinjiang. Methods Feces of obese children in Xinjiang were collected,fecal suspension was prepared,humanized mouse model of intestinal flora(HUM)was prepared,healthy mice were set up as control group(GF),and two groups of mice were treated with GC/MS. The plasma metabolites were studied and PCA and PLS-DA statistical methods were used to find out the characteristic metabolites and identify the characteristic metabolites. Results 183 variables were finally obtained. The PCA,PLS-DA and VIP analysis finally obtained 21 characteristic metabolites;structural identification found that the citric acid,succinic acid,malic acid,etc. of the tricarboxylic acid cycle intermediates in the HUM group were significantly higher than that of the GF group;The expression levels of fructose,pyruvate and oxaloacetate were significantly higher than those of GF group. In addition,this study also found that hippuric acid,such as piperidine formate and 5-aminovalerate in the HUM group were significantly higher than the control group. Conclusion The intestinal microflora may affect the activity of three carboxylic acid and glycolysis in Kazakh children,and the activity of metabolites of intestinal flora is enhanced,and the intestinal nutrient absorption can be promoted,which leads to the obesity of Kazakh children.
