摘要
目的研究线粒体分裂因子(MFF)在肝癌转移中的调控作用。方法利用免疫组化实验,检测12对原发灶组织与转移灶组织中MFF的表达,以明确肝癌转移过程中MFF的表达变化;下调MFF表达后,用细胞划痕实验检测对肝癌细胞迁移的影响;下调MFF表达后,用Transwell实验检测对肝癌细胞侵袭的影响。结果肝癌转移灶组织中MFF表达明显高于原发灶[(0.70±0.06) vs (0.43±0.04)],差异有统计学意义(P<0.05);下调MFF可明显抑制肝癌细胞的迁移[siCtrl vs si-MFF-1 vs si-MFF-2=(77.85±3.65) vs (48.32±1.53) vs (45.32±2.76)],差异有统计学意义(P<0.05);下调MFF可明显抑制肝癌细胞的侵袭[siCtrl vs si-MFF-1 vs si-MFF-2=(29.31±3.53) vs (18.72±2.86) vs(17.68±2.03)],差异有统计学意义(P<0.05)。结论 MFF表达在肝癌转移过程中显著升高,促进了肝癌细胞的迁移与侵袭,提示MFF分子是潜在的肝癌治疗靶点。
Objective To investigate the role of mitochondrial fission factor(MFF) in the regulation of metastasis of hepatocellular carcinoma(HCC) cells. Methods Immunohistochemistry analysis was applied to evaluate the expression levels of MFF in the 12-paired primary and metastatic tissues of HCC; the effect of MFF knockdown on the migration of HCC cells was assessed by the scratch wound healing assay; the effect of MFF knockdown on the invasion of HCC cells was assessed by the transwell invasion assay. Results The expression of MFF in the metastatic HCC tissues was significantly higher than that in primary HCC tissues: primary tissues(0.43±0.04) vs metastatic tissues(0.70±0.06),P〈0.05. The knockdown of MFF significantly suppressed the migration of HCC cells: siCtrl(77.85±3.65) vs si-MFF-1(48.32±1.53) vs si-MFF-2(45.32±2.76), P〈0.05. The knockdown of MFF significantly suppressed the invasion of HCC cells: siCtrl(29.31±3.53) vs si-MFF-1(18.72±2.86) vs si-MFF-2(17.68±2.03), P〈0.05. Conclusion The expression of MFF is significantly elevated during the metastasis of HCC, which promotes both the migration and invasion of HCC cells, suggesting MFF could serve as a therapeutic target for the treatment of HCC.
