硫辛酸对烧伤家兔炎症介质和氧化应激的影响 被引量:6

Effect of lipoic acid on inflammatory mediators in burn rabbits during stress period

摘要 目的观察硫辛酸对烧伤家兔的血清中肿瘤坏死因子-α(TNF-α)、白细胞介素-8(IL-8)、白细胞介素-6(IL-6)和丙二醛(MDA)含量的干预作用。方法在家兔背部两侧造成2处直径为3 cm的圆形烧伤创面,建立深Ⅱ度烧伤新西兰大耳白兔模型。将模型兔随机分为2组:模型组与实验组,每组10只。模型组给予0. 9%NaCl;实验组给予硫辛酸40 mg·kg^(-1)(用0. 9%NaCl溶解硫辛酸),均灌胃给药,每日1次。待家兔创面愈合后,停用药物。分别于烧伤前和烧伤后第1,3,5,7天从家兔耳静脉取血,用放射免疫法检测血清中TNF-α、IL-8与IL-6的含量;用酶联免疫吸附法测定丙二醛(MDA)含量。结果在给予硫辛酸5 d后,实验组与模型组的TNF-α含量分别为(5. 99±2. 36),(7. 12±1. 95) ng·L^(-1);这2组的IL-8含量分别(121. 92±10. 89),(142. 02±9. 87) ng·L^(-1);这2组的IL-6含量分别为(24. 00±7. 15),(26. 12±3. 11) ng·L^(-1);实验组与模型组比较,以上指标差异均有统计学意义(均P <0. 05)。这2组的MDA分别为(5. 37±2. 60),(9. 16±1. 58) nmol·L^(-1),实验组与模型组比较,差异有统计学意义(P <0. 01)。结论硫辛酸可促进烧伤创面愈合,改善氧化应激反应和炎症状态。 Objective To observe the intervention effect of lipoic acid on the changes of tumor necrosis factor-α( TNF-α),interleukins-6( IL-6),interleukins-8( IL-8) and malondialdehyde( MDA) in burn rabbits. Methods New Zealand white rabbits with deep Ⅱ degree burn model was prepared through 2 round wounds with a diameter of 3 cm were created on both sides of the rabbit's back. The rabbits model were randomly divided into two groups: model and experimental groups,each group had 10 rabbits. The rabbits in model group were gevin 0. 9% NaCl;the rabbits in experimental group were gevin 40 mg·kg^-1 lipoic acid( it dissolved in 0. 9% NaCl); with intragastric administration,1 times a day,until the wound healing of the rabbit. Before burn and at the time of 3,5,7 d after burn,the contents of TNF-α,IL-8,IL-6 were measured by radioimmunoassay; and MDA content was detected by enzyme linked immunosorbent assay. Results After 5 d of lipoic acid administration,serum TNF-α levels in experimental and model groups were( 5. 99 ± 2. 36),( 7. 12 ± 1. 95) ng · L^-1; the IL-8 levels in the 2 groups were( 121. 92 ± 10. 89),( 142. 02 ± 9. 87) ng ·L^-1; the IL-6 levels in the 2 groups were( 24. 00 ± 7. 15),( 26. 12 ± 3. 11) ng·L^-1;comparison between experimental group and model group,the differenceof the factors had significantly( all P〈0. 05). The MDA levels in the 2 groups were( 5. 37 ± 2. 60),( 9. 16 ± 1. 58)nmol·L^-1; comparison between experimental group and model group,the difference had significantly( P〈0. 01).Conclusion Lipoic acid obviously promotes the healing of burn by improving oxidative stress and inflammation.
